Karo Question.

About 10 days ago, I knocked up two karo jars. One using a little scrape from a spore print, and another using the inside part of a mushroom stem. The solution inocculated with the spores is crystal clear, and full of whispy mycelial matter. No question, its clean. The one innoculated with the stem, however, is more difficult to decide on. Its roughly 30% full of thick rigourous mycelium, all the little remnants of the stem have huge root like clumps growing out of them, and everything is just very thick.. The problem I have, is the color, and the clairity of the water. Its sort of murky, and yellow. While the mycelium continues to grow. (seems to be getting thicker, and more volumous) The slight yellow color and murkiness began developing about 5 days ago, half way through the mycelium's current growth. I can't find any pictures of karo jars colonized using mushroom stem, so I have nothing to judge from. Is it normal for the water to slowly murk up? When I shake the jar around, it seems like the mycelium is almost soaking in a thin gel, just how it moves, and floats. Its also not so murky that you can't see through it, just not as crystal clear as my spore inoc karo.. Also, when I say its yellow.. Its not YELLOW.. more like the off white color of mushroom stem. The jars have been shaken every other day very vigourously since inoculation. Summary: 1. Should karo jars inoculated with stem tissue and shaken every other day have a slightly yellow-beige tint, and should the water be kind of murky? Anyone have pictures I can compare to? 2. Whats the best way to determine if there are contams in it? Smell? Thanks.

I dont think useing pieces of a stem for karo is a good idea I would think it would cause rot and this could be your case but this is all from my hypothisis

1. no murky is bad. 2. Use the myc solution to shoot up a jar. smelling would just introduce contams.
When cloning try washing the the piece to be cloned in h2o2. Add a few cc's of peroxide to any karo solution that WON'T be used with spore. use a flowhood or a glovebox if at all possible.
