Liquid Culture Contaminated?

SWIM is using Liquid Culture, and isn't sure if it has been contaminated or not. It has been sitting in a dark warm place for 6 days so far, and when SWIM was creating it he was sure to rub down the working area with rubbing alcohol, boil the glass jar in water, rub the lid down with rubbing alcohol before injecting it, keeping the air clean with Lysol while making sure there is no airflow, as well as wearing a mask over his face to prevent breathing on it and flame sterilizing the needle. I would say he took great precautions, but isn't sure if it was successful and doesn't want to waste time with a dead products. (This is SWIMs first time using this method, as well as first time cultivating.) SWIM has provided pictures, which aren't the greatest quality pictures ever. There is smudges on the jar so that is not from inside, and it is hard to see but there is little stuff floating around in there. I don't believe it is fully germinated because there is still black hairs still at the bottom so SWIM needs to wait a few more days still. I apologize if this is the wrong place to post this also, I am new to the forums. And P.S. If it helps, SWIM used honey and pure maple syrup for the sugars. Thanks.

I hope you did all the work in a SAB(still air box), that would help reduce contamination issues. If you did this in open air, then there is a good chance of contamination no matter how clean your workspace is. What you want is zero airflow. Anyway, there is a black thing on the bottom of your first jar that looks weird. Is that a clump of spores? All you need is a few cc's to germinate the LC, not the entire syringe.

I have searched all morning to gather as much information as I could but I figured if I posted pictures of mine as well as my own thread about it I would receive more accurate answers for my situation.

Yes that is a clump of spores, I put about 2CCs in I would say, I still have well over half left in my syringe. And no I did it in open air.

I did not PC it, (I believe that is pressure cook) but I did boil it in water for a half hour, then another half hour and allowed it to cool inside the closed pot.

You us ms syringe? If you did you have to be patient because it can take a little while for the spores to germinate

You should also always PC because the chances you get a contaminate are higher without completing that step, and you waste your time doing all that work for nothing.

Took me 3 tries to get a clean lc using lc to lc and I steam sterilize for 25 minutes

What do you mean, half hour and another half hour. Was this 60mins non stop or what?
I would not use honey in future, unless you are allowing it settle and decanting it off. Proteins come out of solution in honey and it makes it hard to visually see if its contaminated or not. If a clear solution goes milky looking it is contaminated, if its milky to begin with you cannot see this happening.

I microwave most of my LCs for just 20-40mins. When you steam or PC liquids in jars it reaches temp pretty fast, as the liquid is free to naturally move about and mix.

I boiled it for 30 minutes, then put the honey and syrup in the water, then boiled it for 30 minutes more. But the 2nd 30 minutes I turned the heat down to a simmer. I believe this is steam sterilization, but I could be wrong. I followed a guide that is on these forums.

Steam sterilization won't cut it for LC. Also, the chance of contamination from inoculm is greatly increased due to the inoculation of your LC with spores. The educated guess dictates your LC is contaminated.

What recipes and procedures/times did you have failures with while just steaming? if any?
I have never had trouble with microwaved LCs, which will just go a little past regular boiling point. I have steamed/boiled a few too with no trouble, but done far more microwaved ones with no issue. Boiling would take longer as a microwave gets them to boiling point pretty quickly.
I could imagine if trying to do grain based LCs it may need a PC, but I have done honey and maple syrup like the op, at separate times, not a mixutre.

Well, this thread is a good case and point, there, blackout, and you just said yourself that you don't have issues with steam sterilization the few times you've done it when you steam sterilize your LC for longer times, than microwaving to reach boiling point. Guess should have stated that a half an hour of steam sterilization won't cut it with LC. Also, don't inoculate your LC's with spores unless you have sterile spore print taking skills, but that's a whole different story.
