A Noob's Cultivation, Full Tek, +More

Hello hello my fellow Shroomery members!!! Its been a while since I've lasted visited the site...Been busy, as we are all...
But I'm back and with me, I've brought a step by step cultivation tek that many noobs to come, are sure to love!
Over the past few months I have been gathering research and putting together a full presentation, for a full cultivation tek, from when you start rinsing your rye berries, to when you are done drying your harvest.
But yes! Everything! Substrate prep, Jar prep, Tub prep, Casing Prep, Sterile information, an easy Drying Tek, and everything in between.
I'm calling this A Noob's Cultivation, for the specific reason that I intend to explain everything as easy as possible.
----------------------------------------------------------------------
I also want to make sure you are getting the stuff you need without spending a lot of $$.
-----------------------------------------------------------------------
I want to make some of the Teks out there easier for others to understand, and I just want to mainly help out any of those who have questions.
-----------------------------------------------------------------------
As a noob who came into this with only a dream..I would say I am very successful, and I want to share my success with the world
----------------------------------------------------------------------- I promise if you follow the things I am about to show in the thread. You will have a 100% success rate for an end goal of obtaining mushrooms. This means, that if you follow this tek to a T, I guarantee you will eventually acquire mushrooms.
-----------------------------------------------------------------------
You will still run into problems along the way such as, contaminated jars, contaminated cakes, contaminated casings/tubs.
If you are running into more then ONE contaminated jar/casing/tub/cake a batch, you might want to consider the sanitary environment of the area you are working with.
 -----------------------------------------------------------------------
I've been waiting a while until I've been able to get my hands on a good camera to take pictures...and now I finally have that camera.
-----------------------------------------------------------------------
I would like to give a lot of credit to the dedicated members of this site who have shared their knowledge and answered my questions. Now I would like to do the same for others.
-----------------------------------------------------------------------
I am currently taking pictures and getting everything set up. Such as pictures of the pressure cooker and how I use it, pictures of the soils and stuff I use, pictures of the tubs, light set up, etc..
-----------------------------------------------------------------------
-Over time I was able to get some pictures of my progress, but mostly it will be all pictures of things that I have now.
-Photos of some of my first grow-

-Still it will be a step by step presentation followed with pictures and the works. Hope you guys enjoy it.
-----------------------------------------------------------------------
-Only now am I starting to get into strain isolation and such, now that I have mastered my Tek.
-----------------------Stay Tuned---------------------------------

An overview of what you will need.

Most of these things can be found at Wal-Mart, Whole Foods Market, and Home Depot. For the other things.. I will break down what everything is and the prices for the items.
ARE YOU GUYS READY? THE MOMENT ALL THE NOOBIES HAVE BEEN WAITING FOR!!!
-Well, HERE IT IS!
-----------------------------------------------------------------------
1. Jar Prep & Rye /BRF Tek
-I follow TranscendingLife’s jar Tek. Synthetic filters discs with regular mouth jars, are what I use.
The jars I use are 4oz, 8oz, Pint, and Quart size jars.

I clean them off after every use with 70% Alcohol.
(Not 91%, sorry for the photo, will be sure to update with a new one once I get more supplies)
-I suggest ordering SFDs your anything else. It will save you a lot of trouble.
-------------------------------------------------------------
Rye Berries
-I use 4 cups of Rye Berries per bowl and 2 bowls per Pressure cooker. So 8 cups of Rye Berries total. This usually allows me to load up 15 pint jars.
Once the water is clear, I leave the Rye Berries into my Pressure Cooker/Plastic Bowls to soak for 16-24h.
I also add a 1/4cup of gypsum to the water while it soaks, stir it up real good.
(Often I’ll find my pressure cooker is being used, that’s why I also use bowls)
-After 16-24h I bring the Rye Berries to a boil and then drain them. Then I let them dry for no less than 4h but no longer than 8h. After that I load the Rye Berries into the jars and put them into my Pressure Cooker.
----------------------------------------------------------------------
BRF Cakes
I follow the instructions of RogerRabbit, with a few of my own modifications.

I mix gypsum, verm and flour all in a bowl.

I stir it around real good and then add my water. Then stir it up again, load it into the jars, and then put them in the pressure cooker.

Jars will often be messy after you load them. Be sure to wipe off the edges so that the rings where the jar lid sits are completely clear.
)

Once you have wiped them off, put the lids on and throw them bad boys into a PC.
-----------------------------------------------------------------------
2. Pressure Cooker Sterilization

This is the Pressure Cooker that I use.
Fill up the pressure cooker with the proper amount of water, when you’re ready to sterilize the jars. Read the instructions on how much water your pressure cooker needs. Mine only uses 3 quarts while sterilizing.

-I use small jars (4oz) on the bottom of the pressure cooker with a strainer on top of the jars.
-This keeps the jars you are trying to sterilize out of the water and ensure that they are being sterilized with the steam. I have a much less contamination rate when I do this. Only 1 out of every 100 jars or so of mine are contaminated.

-This also allows you to not take up so much space in your pressure cooker while ensuring sterilization. After you put the strainer on the first set of jars, load it with your pint jars. You can usually fit about 10 pint jars on the first level. Once I'm done with that, I put another strainer on top of the pint jars and then load my 8oz and 4oz jars. Total I can typically fit 20 jars In my pressure cooker at once.
-Boiling water only reaches 212 F. To kill off fungal bacteria you need to reach temperatures of 250F+. Therefore, if you have jars that are sitting in the water, the bottoms of those jars are only reaching 212F, meaning you’ll most likely acquire contaminations in those jars. Whereas, if your jars aren't touching the water at all, (sitting on top of the strainer) this means they are reaching temperatures of 250+, a 99.9% success rate, at least these are my results.
-I also prefer smaller jars when I don’t feel like waiting for things to take forever to fruit. The small jars(4oz) usually take the least amount of time to colonize and start fruiting.
-They usually only take about 2-3 weeks from the time you inoculate it with spores, to the time it starts fruiting/pinning in the jar.
They are amazing for stacking in a P.C. (can usually fit 35 or so) and great for those who are impatient
-----------------------------------------------------------------------
3. Room Sterilization

-This is my Air Filter. It is cheap and it does the job quite well actually. PLUS it has another use which I will get to later on. This will literally be your best friend, I know its mine. ^^
-Thats right, I don't use a Laminar Flow hood or anything along those expensive lines...Just an air filter in a small room!

-Place it in a small room (closet, bathroom) and let it run at Speed 2 for at least an hour before you inoculate your jars.
After the pressure cooker has cooled down a bit, (after about 6-8 hours) I place it in the room that I will be inoculating the jars in.
-While inoculating jars, switch it to Speed 3. I do this just for comfort reasons. ^^
I Also spray the room down with disinfectant. Let the room clear out for about 15 minutes before you enter. (This is when I take my shower and get everything ready)

Now you also have an air filter that you can use anywhere in your house! (Multiple uses! Yay!!)
-----------------------------------------------------------------------
4. Personal Sterilization Before you inoculate your jars you want to go through some personal sterilization as well. I take a shower, brush my teeth, use mouth wash, put on NEW CLEAN LAUNDERED CLOTHES, put on my surgical mask and gloves, a tyvek suit and then you’re good to go. (can never be too safe) =)

-----------------------------------------------------------------------
5. Inoculating jars/Incubation
After you have followed Steps 3 &4, now it comes time for the inoculating.
Open up the pressure cooker and take the jars out. I wipe them down with a dry paper towel and then a paper towel that has been soaked in 70% Iso Alcohol.
I flame the needle until it is red hot, then inoculate your jars! (Or bags in this case)

-----------------------------------------------------------------------

Place them in a nice warm area. Try not to let the temp. go above 80 degrees and below 68 degrees.

----------------------------------------------------------------------

-Notice I also have my casings in there too. Getting ready for fruiting!!
Once you start to see signs of growth, then introduce light. 12h on 12h off is what I try to stick too. A little over or under isn’t going to hurt anything. 10h on 14h off or vice versa, Ya know.

-----------------------------------------------------------------------

6. Tub Preparation
Monotubs: For my Monotubs, I follow Damion5050's Tek with a few added modifications of my own. I strongly suggest getting your hands on some Coco Coir before you start. Vermiculite and Peat moss based tubs tend to not let water spread throughout the mycelium that well.

-I've notice that with the Coco Coir, I've had a lot less dry spots, while my mushrooms are fruiting, and during misting, I've found that puddles tend to not gather up on top of the casing/bulk.
I've had the opposite effect when I tried to use Verm/Peat moss for the casing/bulk substrate material.
-My personal modifications are : I use a Peat moss & Perlite organic compost with the Coco Coir. Where Damion says to add vermiculite, I used the Peat moss and Perlite compost.

-The organic compost does contain small amounts of vermiculite as well, but its no solely based on vermiculite. The amount of vermiculite I use is fractional compared to others.
-I also added gypsum, which is one thing Damion doesn't add.

This is gypsum whole.

This is gypsum ground up. (With a coffee grinder) _______________________________________________________________________

-Coco Coir, then Organic Compost, then Gypsum. Close it up, boil up 4 quarts of water and then add it to the tub.
-Let it sit for 8 hours, stirring it up every so often, and you are ready to use it in your tubs/casings!

-Both tubs, as well as all of my tubs always have bubble wrap over the tops to help promote fruiting and retain moisture.

Tub of Golden Teacher that I did. This yielded a combined total of about 5 ounces dry, after the 3rd flush.
-----------------------------------------------------------------------
-Casing Tubs: Same as my monotubs and similar to my cake tubs. I just put all the casings in one tub and mist them a few times every day.

-You can line the bottom with perlite if you want, sometimes I do but more then often I don't. This is only because my tubs usually only fit about one tin case.
-Sometimes I use circular tins and plastic bowls for casings too. When I use these, I am sure to line the bottom of the tub with perlite.
-Cake Tubs: RogerRabbit's SGFC Tek. Ty kind sir for all your knowledge.

The big hole is where I had a humidifier. I don't use those anymore. I stuff the hole with poly fill.
-----------------------------------------------------------------------
7. Casing Preparation
To prepare my casings, I use the same tek I use for my monotubs.
Coco Coir, Gypsum, Peat moss/Perlite combo compost.
The Verm/Peat Tek : I've learned to move away from this tek because I've found that the water doesn't spread through the soil to the mushrooms as easily as it does with the Coco Coir tek, which can be used for casings and tubs.
This Is just my personal choice to use my method mentioned above.
Here is RR's tek on casings.
Strongly recommend purchasing his video as well. He has a lot of creditable and knowledgeable information.
-----------------------------------------------------------------------
8. Fruiting
Air Pump: I've found that I often didn't have time to constantly be fanning out my tubs... so I invested in some air pumps, to keep air flowing for me 24/7. When I added the air pumps to the tubs I noticed a hugeeee difference in the size of the fruits.
-Typical air pump will cost you $10, at Wal-Mart, (can be found in the pet section) and will definitely show its worth in time. The larger air pump will cost you about $30 and can be found at Petco.
-- Here is what my set-up looks like --

--Be sure to clean off your air pump every month and replace the tubing. Water will build up and create mold and stuff within the tubing if proper cleaning is not maintained.

Humidifier: These are optional. The humidifiers I was using tended to make things a little messy and sometimes left this unusual white residue behind. Threw it away after a month of using it.. I got into the habit of just giving them a hefty misting in the morning, afternoon and nighttime. If you want to try and use a humidifier it will just take some time to figure out what works for you.

Misting: The only annoying problem I’ve ran into with misting…is that the bottles constantly break on me..Now I dunno if its just me..but yea I’m buying bottles like once a month. They are only 99cents or so… but really, it is worth it in the end. This way you can monitor your water levels. With a humidifier I would constantly have over saturated mushrooms at times and splitting caps everywhere. Not a problem with the bottle sprayers.
-----------------------------------------------------------------------
9. Drying
Ok, so remember that Air filter we used earlier?
-We’ll guess what? ITS AN AMAZING DRYER TOO!

-Put it in a box with your strainers and mushrooms.

Put it on Speed 3 and let your mushrooms dry for 24-36 hours. Come back and check on them and they should be cracker dry. I REPEAT, CRACKER DRY! That’s right, the envied cracker dry that everyone wants.

-----------------------------------------------------------------------
-If you have some big bad boys though, be sure to split them down the middle of the stem so that they dry out easier.
-Also be sure to keep your mushrooms on the upper layers of your strainers. Otherwise the moisture is just sitting on top of itself.
 -----------------------------------------------------------------------
No desicant, No Epsom salt, no crazy drying non-sense teks, just an air filter in a box with strainers. HOW COOL IS THAT?!?! 24-36h is usually the least amount of time it takes to get them cracker dry. But still, you don’t have to waste your time with all that other stuff. This is A LOT EASIER!
-----------------------------------------------------------------------
10.Enjoy!
Enjoy your dried beauties! Be safe and have fun!

Thanks to all members on this website who have helped me with my previous questions and such. And thanks to all the members who have dedicated their time to help others with their growing techniques.
I hope this helps noobies out a lot!
-----------------------------------------------------------------------

=======================================================================
=======================================================================
=======================================================================
Now lets be clear about one thing, even though this is a %100 success rate that you will eventually acquire mushrooms if you follow this tek...
You will still come across contaminations along the way.
This ranges from :
Jars, Casings, Tubs, Cakes, Agar Dishes, LC and everything in between.
Even I run into a contamination every once in a while, doesn't happen often, but it happens.
Be sure to dispose of these infected products IMMEDIATELY!
For possible contaminations, please refer to Tmethyl's post on contaminations.

--This is Metalobites--
Don't worry if your jars start to show a few of these. Just monitor your temps better ^^
=======================================================================
=======================================================================
======================================================================= -My Challenge To You-
-Through many controlled experiments of mine, I used four different cultures in two different Monotubs. (With multiple trials)
One monotub (Tub A's) contained just peat/perlite/coco coir/gypsum, and the other (Tub B's) contained peat/perlite/coco coir/gypsum/nitrogen.
-Through my observations and studies I've come across a pretty interesting discovery. Tub B's, the ones that contained the nitrogen, was fruiting in-between 1-2 weeks before Tub A's. ALSO, after I harvested Tub B's, I would soak it for 18h (a procedure I apply to all of my casings/tubs/cakes) and by the time I checked back on it 18h later, it would already be pinning again. Tub A's would take 18h to soak, and then for at least another 18h I wouldn't see any signs of pin heads. Tub B's would be covered in .5’-1’ of pinheads across the casing/bulk substrate.

If you intent to try this experiment as well, be sure to grind the nitrogen fertilizer up an and add it to you substrate before you pasteurize it. Pasteurize it, then mix up all your grains with your bulk sub. Stir it up real good and record your results.
When I say pasteurize it, I mean this.
This is the tek I follow now instead of Damion's tek. Frank is another Trusted Cultivator with a lot of creditable and knowledgeable information. I definitely recommend his other teks as well for more advance techniques. ----------------------------------------------------------------------- -Please share your results as well-
As I'm sure we do not all use the same cultures.
I am very interested in seeing different people's results. I'm sure other people would be very interested in seeing these results as well. ----------------------------------------------------------------------- This experiment was only preformed with 4 cultures TC, GT, B+(crap culture IMO) and PES Hawaiian.
I do not know if you will have the same results with other cultures.
I intend to eventually use this different tek with multiple different cultures to test the validity behind this experiment.
Gl if you choose to try it.
-I will re-conduct this experiment with 4 new cultures and one of the previous cultures and provide pictures to show the proof, if any;).
----Pictures of this soon!----
-----------------------------------------------------------------------
Hope you all enjoy the tek.
Everything from here on out will be all of my updated progress. A lot of it is experimental work, so be sure to stick to the things that were provided above unless you want to get into some crazy work.
If you want to get a little crazy and experimental, be safe! Make sure nothing you are doing could potentially poison/kill you! RESEARCH AND READ ALL THE INFORMATION THAT IS AVAILABLE TO YOU! It could save your life.
Also, allow me to make some mistakes for you. I am very careful about my work to ensure the safety of not only myself, but everyone else as well. I want to help everyone out, but I also want to learn some new things.
Enjoy your stay, hope to see you on the forums!

Off to a bad start but good improvements. I can see in your picture you are still using Permanent marker. Expo ftw?

Clean and reusable jars? Maybe its just me..but I like to reuse jars and often I don't find myself using the same culture in the same jars. When you start to have several different labels in permanent marker, things get messy and confusing.

u can just remove the permanent marker easily with the expo marker remover, or some cologne or ur girlfriends body spray (most of those things are based with denatured alcohol) The marks usually come right off.

I've found this to not be the case with permanent marker..You can use the expo trick..but why not just use the expo in the first place? If you want to waste your cologne/girlfriends body spray, thats on you. I know I would get called out if I used my girlfriends perfume.. A 2 pack of expo markers is $2 dollars at walmart. Money and time well spent.

Lol..the point is..why take that extra time i guess..if you have time to wash 20 jars one by one cool. I prefer the simple wiping off.

I wash hundreds of jars one by one every month. It's one of the many fascinating and exciting elements of my job
When I have 200 jars of different strains and species laying around all looking pretty much the same, I can't have my marker just rub off when it feels like it.
Also I make sure to wipe my jars down with isopropyl before going to work with them doing inoculations or grain to grain. There is absolutely no chance anything less than a permanent marker will survive that
I've gotten confused plenty enough on what jar is what after the marker gets rubbed off accidentally so that is why I need a sharpie. It works great for me

I have recently cleaned out and cleared out my whole grow room to make room for a new grow and to start over with my new knowledge of the subject.
For starts, I will be using Wild Bird Feed in place of Rye Berries.

I followed the same rinse procedure that I used with the rye berries, soaked them for 18h(with gypsum)
-Brought the bird seed to a boil, loaded my jars and then loaded the pressure cooker. I cooked it at 15 PSI for 1h30m and let it cool for 8h.

I inoculated the jars after everything cooled down.
(Jars inoculated on the 10th)
This is a result of 10 days later.

And these are some BRF cakes I did on the 4th

These were all multi-strain batches.
--Pictures of strain isolated batches to come--

1. My first attempt at isolating strains so that I only have Rhizomorphic strained cultures!
-First I took a Petri Dish with and inoculated it with a multi-strain spore syringe. I let it sit for about 2 weeks and this is what I came back to. Should took pictures before I started cutting lol. Didn't want to risk the contamination possibilities though.

-Then I took a knife, flame it red hot and cooled it off with a paper towel soaked in 91% alcohol. I then cut out pieces of the agar that resembled ice sickle like structures. This is what I got 2 days after my first transfer.

This is the growth 2 days after that.

I will be preforming my 3rd set of transfers in a day or two.

_______________________________________________________________________
Edit, Update 5/22
Got my hands on some Karo.

Waiting on my Petri Dishes to arrive so I can preform my third set of transfers. After the third transfer I will decide if the strain is isolated to the desired point.

From there I will either preform a 4th set of transfers or start liquid cultures.
My hopes are..
With the successful isolation of the rhizomorphic strains with my different cultures...
and with the use of liquid cultures as a mean of colonization for my bulk substrate and cakes...
I can get the colonization time on my cakes and substrates cut in half.
-----------------------------------------------------------------------
Though this is already a given through what mycologists have already discovered and told us...
-----------------------------------------------------------------------
I plan on using these liquid cultures mainly with my smaller (4oz&8oz) jars.
These results will hopefully show as :
4oz - Jars take 7 days maximum from time of inoculation to when it pins in the jar.
8oz - Jars take 11 days maximum from time of inoculation to when it pins in the jar.
-After the maximum amount of days have been reach, I'm hoping by the 3rd day, both of the cakes will be covered in full sized, glorious mushrooms.
-Both Jars will receive 5cc of liquid culture-
-----------------------------------------------------------------------
With the cultures being strictly rhizomorphic strains...
I'm hoping to yield :
4oz - 1oz dried, combined total, after the 3rd flush.
8oz - 3oz dried, combined total, after the 3rd flush.
-----------------------------------------------------------------------
Of course, this will increase the rate at which my pint jars colonize as well. As well as improve the quality and size of the mushrooms they provide.
My hope for results are :
Pint : Jars take 14 days maximum from time of inoculation to when it pins in the jar.
Pint : 6-8oz dried, combined total, after the 3rd flush.
Jars will receive 5cc of liquid culture _______________________________________________________________________
-Stay Tuned-

I wouldn't bank on that
