Liquid Growth Medium
    #1550965 - 05/15/03 12:10 PM (20 years, 11 months ago)

I've used the Liquid Growth Medium tek to innoculate many jars from a single spore syringe.

I thought I had another prepared syringe in my refrigerator, but I was mistaken and was wanting to innoculate a few more jars this weekend.

I can't think of any reason why I can't just add established mycelium to my powdered malt extract solution. Has anyone tried this with success and if so, could you please offer a few tips?

Thanks, in advance.

even better, using existing liquid cultures
to innoc subsequent batches.

less time waiting for spores to germinate.

but it is worth noting that doing that is similar
to grain to grain transfers, in that any latent
contamination (invisible or otherwise) will
be transfered from the old batch into the fresh
new batch.

in short, make sure the source is clean.

Just watch out -- if you do it too many times the strain will degrade (esp. if using the same media each time.) S'why petries are better -- you can actually look at the mycelium and sector rhizomorphic parts which will prolong the strain's good characteristics instead of taking random parts.

Thanks so much, afoaf .

I periodically use a Sterilization Test Kit, but only when I work with bulk teks. I know it's not 100% effective, but it gives me a little more confidence against contaminants.

If you want one just look up Sterilization Test Kits on any search engine. A friend of mine owns a body piercing studio and told me about them.

They are usually about $5 a test and are well worth the alternate aggravation of lost time and effort.

I've also been using bacteria water test kits. They are meant for people with low immune systems to test water for harmful bacterias, but are great for testing moist substrates as well. A cheap bacteria test kit can be found online or at larger pharmacies for around $20.

Thanks again for the info, afoaf. See you around, pal.
