Agar's GLC Tek *click me* 4

I'm just posting a link of Agars glc tek on the culivation forum because its so helpful. To all noobs looking for a good way to multiply culture. Here it is. http://www.shroomery.org/forums/showflat.php/Number/6817701/page/2/fpart/1
Every time I order a new strain I just squirt a tiny bit of spore solution into a glc jar per this tek and that alone gets you around 500cc of 'glc' of that strain. Which is about 40-50 syringes. Then you still got a whole syringe full of spore solution which looks like nothing when the glc colonizes.
Need a PC
Tek Quote:
agar said:
Pint jar on right is multispore inoculated (SA). Allowed to colonize near 100%. Syringe is 60 ML w/18 gauge needle. Quart jar on left is sterilized water.
Aspirated 60 ML sterile water into syringe.
Injected 60 ML sterile water into colonized grain jar.
Shook grain jar to liberate mycelium from grain.
Injected total of 180 ML sterile water into grain jar. (shaking mildly between each injection)
Aspirated out mycelium laden water from grain jar. (all I could aspirate out of the grain jar was 150 ML) Then, injected mycelium laden water back into sterile water jar. Approximately 500 ML of Mycelium laden water. (enough LC for approximately 50....10 ML syringes)
The advantage of this procedure is that you can be assured. The culture in clean. Because you can VISUALLY inspect colonization in the grain jar. (if there is FUNK, you can see it )
That is not the case, with sugar type LC's. (multispore syringe inoculated) They can be contaminated, from day 1. And, you NEVER know it. Until you inoculate spawn jars with that LC. Then, have every single spawn jar GO SOUTH.

Ya and u can use silicone to blob over the injection hole so you don't need to go all fancy with proffesional ports like Agar used. Simply tape tyvek over a 1/2" size hole with micropore tape as the filter. You can make the port far on one end of the lid and the filter far on the other end so you can lay the entire jar down on its flat side and water won't touch the filter. You can then stick syringes in the port one by one aspirating each one full of the 'glc'.
All of this is performed in a still air box and the syringes are pre pc'd and freshly unwrapped inside the glovebox before use.
I blob the silicone on the bottom of the lid and let it sink thru the other end so I can see it easy for the needle. I'm sure you can blob it on top but I just found it hard to see where the hole was at every time.

How long does the glc remain viable ? Should I store it in the fridge ?

Usually around 5-6 months when stored under refrigeration.

Is the jars you got the myc worthless now or will it replenish?

Most of the time the grains are left so wet you would just wanna throw it away. At least I do. I just inoc more glc with the glc lol and by the time you run out of your first batch you'll have more.
This tek is the shiznit
