Agar Transfers

Ive heard fairy tales of people having major contamination on agar plates and rescuing the mycelium from one plate to another. ok so here is my situation
I know it wouldnt be worth it, but i have run out of cultures and this is the last little peice of my strain that i have left so, im desperate. Any suggestions would be appreciated

All you can do is to transfer several small pieces from the center to fresh peroxidated plates and hope for the best. After transfer check the plates frequently for new growth and as soon as you see it transfer again to new plates.
I don't see exactly what you have there - if you have only bacterial contamns then your chances are good. Good luck!

ok heres another problem. No i dont want a "your Hopeless" but all ihave right now (no money) are Three DFA plates and a bottle of peroxide. Is it possible to do this without peroxidated agar? and in only three transfers?

BTW what you heard are not fairy tales Especially when you try to germinate spores collected from wild you get such plates.

It's possible to do it in three transfers but it will be harder without peroxide. Transfer at least 4 chunks to the first plate and hope that at least one will work - you got nothing to loose. Good luck.

You could wash the mycellium with few drops of peroxide or spray it on the agar surface (if your agar isn't peroxidated), but I didn't have much success that way. Maybe someone else could give you an advice on this.

dump your plates into a jar, remelt them and add some peroxide, pour 3 more plates.

Solidified agar will still hold additional fluid (water/peroxide/GS). Simply add .25-.5 cc of 3% peroxide to the surface of each plate. Allow the peroxide to be absorbed and then transfer to the plate. Make sure to allow the peroxide to form a thin layer over the entire surface of the agar plate.

IF I am seeing correctly you have a case of red or pink sploches in those cultures ? This could be either Neurospora , or maybe Trichotecium ,and if so is best that you not try to salvage the mycelium . Each time you try you may actually make things worse . You could , if desperate try using some gentamycin sulfate mixed into a batch of agar , then trasfer some of these risky cultures to that . IMHO You should "S" can these and start again , better safe than sorry . Good Luck

No theres no red or pink (thank god) thanks joshua i think ill try that how long should i wait and how do I avoid condensation?

Must be my monitor , or your pic , sorry bout that . It is no fairy tail though you can succesfully salvage a culture , in three dishes .....that is cutting yourself a little short . Just harvest the best , as far away from the contamed area as possible . Make sure you move very methodically , being carefull to not disturb the contaminated area even with a tiny little draft of air . Keep an eye on those new cultures , and repeat as many times as you need to to clean the culture .
