Agar culture highly contaminated
    #1409344 - 03/25/03 03:55 PM (20 years, 11 months ago)

I just started a culture a few (3) days ago and its contaminated (ants got around the edges due to my shitty sealing job). I was wondering if i can keep it and wait for mycelium to grow more, then cut it out out and try to transfer away the contaminates. Or, just toss it out now and start over. It hasnt gone for very long, so that wouldnt be too big of a problem.

If its contaminated....I'd start over.

Stick your cultures in a sealed container...keeps bugs out.

Tip :
You can use black electrical tape to seal the petri dish.

good luck!

Hey roadkill if your willing i have a few more questions about agar cultures
1. Best temp to incubate
2. Have you ever used just dogfood and agar before?
3. Light. Is it always a bad idea in this stage or not important?
4. Thie bigger the piece i throw in there the faster my results or am i just kidding myself?

I incubate at room temp...74 degrees F.
I have wonderful growth at this temp.

I have never tried dogfood agar...and probably won't.
I have a dog and the food...but it seems like making it is a pain in the ass.
I'll stick to MEA and PDYA.

Well if you give the plates light you will have small mushrooms growing on them.
I keep mine in the dark.

Bigger has nothing to do with making agressive mycelium.
If you are cloning...cut out a small piece of the stem and peel the outside portion off...then dunk it in a mix of hydrogen peroxide and water before putting the clone on the agar.

I'll take a picture of a South African clone that I have on Agar now and post it in a few minutes.

good luck!

just lay the peice gently onto the agar.

South African Clone on MEA.

South African on PDYA...1st transfer...this strain is amazing!

As you can see from the pictures the South African is very agressive!!

Wow That is truly amazing!! How do you deal with water condensation it looks to me like youve got none at all.

P.S. Im very jealous of those nice looking petri dishes Pyrex i assume?

I don't get any condensation on my plates.

I have Pyrex and Kimtex petri dishes...and some poly autoclaveable ones that I haven't tried yet.
The poly are expensive...they were given to me by a good friend.

I'm pretty impressed with the South African!

good luck to you!

yah i noticed that right away it was downright nasty. SO made two new plates both with chunks from the inside of the stems and put them in the petris using the oven tek. The plates were sealed using Electrical tape (Thanks ROadkill)
