rubbing alcohol harmful?
    #1392045 - 03/19/03 02:21 PM (21 years, 15 days ago)

AFOAF wondering if anyone makes a solution to sterilize everything they'll be working with when making prints, syringes, birthing jars, etc. would a tub with bleach water, or a tub with alcohol, or maybe h202 water, would any of these work, the friend is really worried about contamination, which im sure everyone can relate to. he'd just like to know if something like this would work, also does rubbing alcohol harm spores, diluted or otherwise, sometimes he has remnants of it on his hands/gloves. he's read the articles on sterile techniques, these are just some questions he didnt see and would really appreciate any help you can give. thanks a shit load in advance.

anything that kills germs will kill spores. a good thing to disinfect with is a 10% bleach/water solution.

for the actual surface you'll be printing on, that should be heat sterilized.

so if he sterilizes a shot glass, a scraper, and all that in bleach water, using the oven draft method, then he should let the heat dry these things out, correct?

Bleach water will leave a residue, especially if you soak something in it for over 30 minutes. For spore printing, heat sterilize your paper/etc. in an oven covered with a drinking glass. Put a bottle of rubbing alcohol in a spray bottle and spray the shot glass -- the alcohol will evaporate leaving no residue. SWIM uses alcohol for everything -- hands, arms, containers, etc. Just let it air dry -- you will feel your skin cool as it evaporates. SWIM says alcohol rules -- he has had a *very* low contamination rate using rubbing alcohol .... just let it evaporate and you will be fine.

this may seem a little complicated, but you'll get good sterile prints:

get a peice of aluminum foil that's a good size for the print. put it in a jar with a filter on it and PC the jar for an hour at 15 PSI. (if you don't have a PC, steaming the jar should be fine). the foil should be on the bottom of the jar. it's helpful if you've got a glovebox for some of this, but it's not totally necessary... when the jar cools and you've got a nice planar-capped mushroom growing, you're ready to print. go into the cleanest (and preferably smallest) room in your house. (i'd suggest the bathroom). bring your terrarium in there and bring your printing jar in there. spray lysol liberally in the air. open the terrarium and loosen the lid on the jar. don latex gloves and swab them with alcohol. with a pair of flame-sterilized scissors, cut the cap close the the stem. hold the cap by the edges, don't touch the bottom. with your other hand, take the jar's lid off, and quickly put the cap on the foil in the jar. then put the lid back on... all very quickly and smoothly. after 24 hours, observing the same glove\alcohol\lysol\clean room procedure, quickly open the jar and remove the print with a pair of flame sterilized tweezers into a never-before-opened plastic bag. viola.

I am assuming either a polyfil or filter disk lid. The kind you would use when preparing grains, but with the tin foil in the jar instead of all that smelly rye

when you flame sterilize, how long do you keep the flame on something, and is it just an ordinary lighter, or should i put alcohol on it and then flame that?

the filter is there because there's gonna need to be air movement in and out of the jar because of the temperature differences that are going to be going on... you don't want contams coming in with that air. there should be a layer of aluminum foil ontop of the jar too to keep water from dripping or splashing on and in. flame sterilize for 15 seconds or so. don't hold the instruments in the flame, but just above it.. it's hotter there and if you hold it in the flame, it'll get sooty.

1 part bleach, 2 parts vinegar, 100 parts water = excellent herbicide. me=lazy so I just use some random combo of water and bleach...probably way too much bleach. My roomate is from hell...there is shit growing in my kitchen that would make you wonder how I dont get food sickness daily. I work in my room (where there are probably spores abound). Spray lysol, allow to settle, do that again, clean all working surfaces with Isopropyl 70 or 91% and use it to sterilize surfaces except use a flame for syringes, scalpels, etc and then wipe with ispropyl soaked towel....I have never PCed anything for "labwork" type stuff and have low contamination rates. See sporeworks' syringe tek for an example of such sterile techniques.....I think most people do overkill.....but not a bad idea for those not used to the cleanliness.

Good luck - MM

Mush master but what does the lid look like is it just the outside ring holding the filter on? Or is it a lid with a hole in it?

you take the lid and punch a hole in it about 3/8 of an inch wide. then take some polyfil and roll it a little and thread it through the hole... you pull it through so it's nice and tight.
