Inoc holes covered during incubation
    #1360412 - 03/09/03 12:07 PM (21 years, 1 month ago)

Hi -
A friend of my monkeys uncle has covered up the jar`s holes with masking tape and placed them in the incubator for three days, no growth was started yet, but as I was looking in the forum and FAQ for answers for him, I noticed a post that stated that the holes SHOULDNT be covered up, is this true? Or is he panicing for nothing?

in the pf tek it says to cover the holes w/ that medical tape , which is like gauze,,,
so that there can be an air exchange ..so it can breathe!

I'd sterilze some doubled-up foil, uncover those holes, and crumple the foil over the tops of the jars to prevent contams from "falling" through the holes to the substrate.

So with masking tape it will be unable to breathe and stall?

Masking tape is not porous.

It does not really matter right after inoculation. As the jars become colonized, removing the tape can help speed things along and prevent stalling.

Phew. Thank you gents. Was a lil worried for him then. Whilst I`m here I also have another question, if anyone would be able to help, it would be appreciated. My friend Tony Blair PM, after sterilising the jars with substrate in, left them for a day in a dark cupboard with the holes uncovered, would this have dried out the substrate?
