Wild Print to Agar

OK, so I understand when you go from WILD spore to agar, you always end up going to at least one or two more plate transfers afterwards to clean up.

But what I'm curious about is... is this done because mold will *always* grow from wild prints or do you just transfer to new plates for good measure?

And if contam is such an issue... how can you even be sure which one is the correct mycelium?

You can probably count on some type of bacterial contam, or mold because they have been exposed to open air for long periods of time.

The correct mycelium doesn't turn green :tongue:
Bacteria doesn't look like mycelium.

Sometimes it's hard to tell what is and isn't healthy. mold will look wispy and has a "moldy" looking texture, if that makes any sense

If you have the wild mushroom, you could rip open the stem and get tissue from the inside, which has not been exposed to air, and put that on agar.

You might not contam that way, so you wouldn't have to transfer away from contams. If you see no contaminations, there are no contaminants, which is the beauty of working on a 2D surface

Yeah man, don't over think it.

You'll spot the contaminants easily, and any contaminant that is white like mycelium will not produce rhizomorphs like mycelium, unless of course those contaminants are other spores!! :grin:

I just started working with agar... is it safe to still take an isolate if there is a contam visibile?  (obviously I wouldn't slice the contam section)

even in RR's videos he tells that he had to take contaminated areas of the large oyster at times to isolate the myc growth. also try to take your slice furthest away from any contams.
one of the things that is also mentioned by people is to cut your mycelinated agar and turn over the piece you want to keep and put it face down on the new agar plate so any contams will have to grow out from underneith--the theory expressed is that the mycelium will "outrun" the contaminates.
after two to four plates it is possible to be contam-free. RR says so.
