incubation problems
    #1028287 - 11/06/02 01:46 PM (21 years, 4 months ago)

On Oct. 27 I innoculated 10 jars with B+ syringe. I am incubating the jars in there original jar box, on top of a heater (check pic).

My problem is that only 3 jars of the 10 have started growth. All jars have 4 punched holes in lids. 9 of the jars have all the tape removed. and the 1 left over has tape on all 4 holes, but this is one of the 3 showing growth.

I dont know if the heater is drying the jars (realitive humidity is around 30%) out or if the fluxating temps is a problem.

I have the jars on the heater at night for about 7-8 hrs, temps are around 84F. I have to go to school so i cant leave the heater on. During the remaining 16hrs of the day the jars are around mid 60s. Does anyone have a possible antidote for my situation?

Just create an incubator with an aquarium heater in a jug, and you'll be fine.

It might just be that the dry heat is sucking up moisture, it could be that its too hot...it could be that the temps are so far away from 86F either too low or too high that is casuing your problems.

It will really make things slow down if your temps are not optimum 24/7

You can try injecting the cakes with sterile water, but this may introduce contams, and it isn't very successful at distributing the water uniformly. Do like angryshroom said and build you one of them nice, cheap styrofoam cooler incubators using a fish tank heater in a jar of water/peroxide solution.

How much are the fish tank heater. like $20 give or take?

and until i get one, can i continue to incubate them the way i have been.. 60F during the day and 84F at night? or just leave them at a constant 60F?

I got mine for $13. I'm pretty much sure that the constantly changing temps aren't great for incubation purposes, so I'd say take them, wrap them up in a blanket, and put them in your closet, go out to walmart and buy a styrofoam cooler and get a fish tank heater asap.
