Mercuric chloride solution for external disinfection of mushrooms

Someone of you knows how to prepare and use a solution of mercuric chloride  as an auxiliary agent used for prior external disinfection of the mushrooms that will be used for isolation of mycelium on agar ?
I have some Auricularias growing in a mango tree at home and I have not been successful in isolating a clean culture of it. Last week, I also received five sun-dried Cordyceps militaris from a friend in Portugal and I want to isolate it also.

Try antibiotic agar - much safer, much more environmentally friendly and effective.

Yes, I know about that. I have mercuric chloride in salt form and I want to test its efficiency with all necessary care. Iodine could do the same effect ? If so , what is the correct concentration of iodine diluted in alcohol ? I have used antibiotic media (Sabouraud agar plus yeast extract,Gentamycin and cloranphenicol) but I am still having some problems with yeasts.

Do you want to do anything with the mushrooms afterwards, e.g. consuming? Forget about it.

Mercury is highly toxic and it accumulates in your body.

Use tincture of iodine. Much safer, just as effective.

Buy it cheap at any pharmacy, already mixed.

Dead is dead.

You can find iodine (or Betadine) pads at a pharmacy, in the first aid section. Individually wrapped, ready to use.

I believe that the tincture of iodine is too strong and can kill the spores of the Cordyceps. The alcohol present in the tincture can kill all (mycelium and Cordyceps spores too).

I tested a weak solution of povidone-iodine (Betadine). The concentration of the commercial solution is 10%. Too strong for direct use.
I prepared a diluted Betadine solution in sterile distilled water (3 to 4 drops of commercial solution in 4 ml of sterile distilled water in a test tube).
First of all, the Cordyceps mushrooms are cut into small pieces (4 mm) with scissors, transferred to test tubes with sterile distilled water, agitated for a few seconds to re-hydration and to make a initial cleaning. Then the pieces are dipped in the diluted solution of Betadine for 10-15 seconds. After that, the pieces are washed 3 times in sterile distilled water . Then the pieces are transferred to an agar with gentamicin sulfate. This is the method that seems to be working well.

No, donĀ“t worry about that. The small pieces of the mushrooms will be only used for mycelium generation . The 1 ml solution of mercuric chloride used for dipping the mushrooms will be left in a Lab with an adequate final destination for the waste.

I discovered that a 0.1 % solution of mercuric chloride is too strong for surface sterilization. Maybe a more weak solution could work well. This salt kill all the spores : the ascospores of the Cordyceps inside the perithecia structures and also any spores of contaminants .It can kill also any viable mycelium inside of the Cordyceps fruitbodies).

Anyway, here is the mercuric chloride method I have used:
