To much condensation under lids of Petr dish
    #10196062 - 04/19/09 03:27 PM (14 years, 11 months ago)

I hope someone can help me out with this question.Ihave had a lot of trouble getting the past 20-40 jars to colonize after inoculation them with 1cc of spore solution.I almost gave up because I had no clue what the problem was I had been growing for some time be for I experienced this problem.
So a few weeks ago I decided to give it another try and I ordered the syringes and had them shipped not even 90miles from the distributor so there would be NO problems due to shipping. When they arrived I made 10 jars and knocked them up,placed them into the incubator, and guess what .i HAD THE SAME FUCKING PROBLEM.But anyways I decided o order a pre sterilized agar kit and knock up some dishes.I knocked up the dishes Fri night and by Sat afternoon there was so much water under the lid I knew this had to be a problem.I wiped them down and checked them again 30min ago and the water is back.Is this a bad thing? And if so how do I prevent this.Any help would be appreciated.
  thank you

The condensation is from a difference in temperature from the outside of the petri dish and the inside.

Just like pulling a bottle of liquor from the freezer. It forms ice all over the outside. The air outside the bottle is warmer than the glass, causing the air to cool around the glass, and condensing on it. The air in your petri dish is getting warmer than the outside of it. Someone please correct my if my physics are wrong.

I'd get it out of the incubator and hang the petri dishes upside down so the water can't run down the edges into your agar.

Sounds like high humidity is the main cause. If you want to lower the humidity in a small incubator....maybe get some of those moisture absorbing things that come in bags of beef jerky and inside brand new shoes. Go to your local payless and just grab a bunch. =P

EDIT: I haven't tried this. Wait for someone else to chime in and either confirm my claim or deny it.

As long as the inside top surface temperature of your dishes is lower than the air temp/water vapor temp within your dishes, and humidity is high enough within the dishes, you'll get condensation.  Warmer air can hold more moisture, so you're best option might be to take everything out of the incubator to reduce humidity within the dishes.  Other things that will work, insulation or heating the tops of the dishes.  A good trick is to use a top down desk lamp.
