Question on Grain Spawn

Hey guys,
So I finnaly got my hands on a pressure cooker and have steralized some WBS that I got and have put them in mason jars with the Polyfill in the lid.

Normaly, I would Inoculate them with a syringe, but I am planing to drop a few Oyster Plugs in, becouse thats all I have.

What is the best and most sterile way to do this? I have to open the lid obviusly, so should I make a glove box, or what?

The most sterile way would be to inoculate in a glove box or in front of a flow hood. Just open the lid and drop them in using some foreceps or tweezers. Depending on where you got your plugs the may not be sterile. I know when I make plugs they arn't sterile... perhaps someone who knows more about plug producers can chime in on this. If you inoculate grain with unclean dowels you are asking for contams.

If they are already colonized sterilizing them will kill the oyster mycelium. If you make your own plugs you can sterilize them or not. Where did you get the plugs from? You could always try to inoculate the grain with them to see what happens. If it contams then you know, if not your good to go. Alternatively if you work with agar you can put a plug on an agar dish and isolate away from contams and once you get a pure culture you can inoculate your grain with the agar.

Could you sterilize the Plugs with hydrogen peroxide? Or would that kill it too?

Yes, you could dip them in H2O2 briefly to kill some of the contaminants off of the surface. I would stick to agar, but you could try it out, Oysters are a pretty aggressive strain. I would personally use iodine or bleach instead of H2O2, and then use a sterile scalpel to cut away a layer off the outside. The brief dip kills some contams, and the slicing opens up the inside so there is faster grow. All of this is IMO though...

with the mycelium actively growing, and contaminants having to start "from scratch" it should easily outrun them. You could us H2O2 or Bleach or iodine, but it would tend to set the mycelium back and lessen it's ability to run right away.

I would just chuck your plugs in.
