Spore syringe help
    #10020829 - 03/22/09 04:14 PM (15 years, 9 days ago)

I'm making a spore syringe, and I need some help

I have:
•2x 10cc syringe
•1 fairly large spore print (from what I've read/seen it seems to be capable of producing about 10 syringes)
•distilled water
•half-pint jar
•shot-glass
•"Sex-lube"
•alcohol burner

I was going to follow the tek from RRs video, but don't feel confident about my sterile procedures and I don't have an inoculating loop. Instead, I thought I might just make a jar full of spore solution with a self-healing inoculation port (then I could just shake the jar and suck up 10cc of solution whenever I needed to).

My tentative procedure:
1. sterilize jar of distilled water in PC.
2. in glove box, open jar and drop print in, close jar.
3. shake vigorously to remove spores from foil.
4. draw solution into sterile syringe.

Would this work? I see no reason why it wouldn't, but I'm a noob.

If it won't work, what could I use in place of an inoculating loop (preferably something I can find around the house)?

Don't drop the print in the jar just scrape some in the jar and the rest should work,  the out side of most prints are not sterile so its a bad idea to drop the print in.

Oops just sterilize a butter knife to scrape the spores off.

don't drop the spore print into your sterilized jar of water with the injection port...
the outside part of the material that the spore print is on, probably has a ton of contaminates on it.

scrape the spores off the print with some kind of instrument, into your jar.

you can make an inoculating loop easily.

or you can use an Exacto knife, just wipe it down with alcohol and flame the knife before using.

Thanks, I have a butter knife that will work perfectly for this.

My plan was, otherwise, sound, correct? I can make a big jar of spore solution?

Assuming everything is 100% sterile, you can do anything. However, that is not realistic. Spores should be stored dry. They keep better that way.

I recommend this:

I'm worried that my glove-box won't be enough to keep the syringes/print sterile. Are my worries unfounded?

I don't want to waste spores.

Another thing.

I plan on using all 10 syringes worth of spores in the next couple weeks or so. Does that timeline make my plan more feasible?

Prints are not 100% sterile in the first place. Just work quickly in the still-air, make sure to wear gloves, and sterilize your tools. Then store your spores away again.

A spore syringe doesn't need that many spores. even a miniscule amount will contain thousands or more. Thus, if you mess up, you'll have more to use later.

Do it as best you can following that video and good luck! :vaped:

the spores will remain viable for a very long time, it does not really matter when you use them. However, some people say they get faster germination from fresh spores. But there is little difference as far as I have read...

Damn. I really thought I might be able to come up with a way to minimize my work and chances of contamination. :frown:

Oh, well.

Would you care to explain how I would be cutting it short? I don't quite understand.

Edit:
Thanks, you've allayed my fears. I now feel confident in following the directions in RRs video.
