thats a good question, I was thinking of cutting a 2 liter pop bottle in two then shitting in it then pissing then taping it back togather air tight then putting the balloon on top.

Mixing the poo and urine till homogenous would probably help the urine activate it. You could break up the poo in a plastic bag, cut the bottom of the bag and squeaze it into a larger mouthed liquor bottle quite easily, then urinate in it, put the lid back on and shake.
I think this would be the best method.
