The toxicity levels of Psilocin and Psilocybin are actually quite low, so there aren't really any well-established long-term physical affects.
In 1961, Leary and Alpert started a short-lived wave of studies with the 'Harvard Psilocybin Project', though studies on hallucinogens and its therapeutic and psychological benefits have gained popularity recently, it hasn't been widely noted as an interested in medical study since.
In the interest of being brief, I wont go into details about the neurological and psychological affects of Psilocin (What Psilocybin turns into in the body) and allow you to do your own research. I can, however, hypothesize that anything, like Psilocin, that has such an affect on the Brain and Neurotransmitters, can have potentially adverse long-term psychological affects.
Psilocybin Mushrooms can be used therapeutically and aid in psychotherapy, but it can contribute to or worsen psychosis and schizophrenia in unregulated dosages.
In other words, I would advise playing it safe and to enjoy the positive affects of this wonderful drug in moderation... and like someone else said... it really is better to do it once in a while, the magic is much stronger.
