Two Lophs just started in culture:

Another Loph after about two weeks:

...and a few months:

San Pedro pupping in vitro:

Starting a bunch of seedling grafts:

Baby Lady slipper orchid:

Poppies grown entirely indoors, it can be worth it! (color is weird making the plants look pale because of my HPS light):

I want to know more about cultures and why they are done and how you do it

You can include sugars in the medium which the plant can use instead of the plant having to photosynthesize it all. It can speed up growth significantly.

Okay, I am definitely, DEFINITELY, replacing my T5 fluorescents with an HPS for indoor poppies. I've never gotten a poppy to that size indoors.

Well that is not all the HPS work. This guy knows how to grow poppies. You can grow some really nice shit with Fluorescent, BUT you have to REALLY know how to to grow poppies for anything that will look good.
Now i do love fluorescent lights. I have seen some Major stuff come from fluorescents that made some peoples HPS stuff look like it grew in a cave.
Gotta know how to treat the plant just right and it will reward you. Fluoro or HPS.
I really prefer a Mix of CMH and Fluorescent side lighting really close in.

Growing plants in culture is a pretty expansive topic and the procedure is different for every plant (and completely dependent on what you're trying to do!). I'd be happy to answer any specific questions you have about tissue culture, as best I can, but I wouldn't know where to start with a question so vast as "How do I grow plants in culture?"!. There is a good book for starting out if you're seriously interested called "Plants from test tubes" that I'd recommend, you might also do some googling - "micropropagation", "plant tissue culture", etc.
There are many benefits of growing plants in culture, for one I like the challenge! Not to mention the ability to clone difficult or impossible to clone plants (by traditional means) rapidly. You're also able to grow slow growing plants very quickly, which is obviously an advantage, I do lots of grafting to and with my lophs the growth rate in culture is ~3x that of a plant grafted to a persk.! The other benefit of cultures is that they don't need much light at all (actually they can be grown in complete darkness technically, but this isn't usually done for various reasons, all the sugar needed for growth is in the media) or really any sort of care, I don't need to water my plants in culture or deal with bugs or anything like that. It's a great way to store/keep stalk of valuable plants you might have, and many plants in culture may even be put into a fridge etc. for long term storage without damage! I'm sure I could think of many other reasons to get into plant tissue culture but I'll spare you my ramblings!

Saliva tissue culture does work, although I've never been able to get a proper callous despite much effort! Nodal cuttings work great without hormones in MS media with vitamins and 3% sucrose. It can be a little tricky to get a culture established, they can be hard to sterilize without killing the plant but once you've got one started it's easy. Although Salvia is so simple to propagate/grow by normal cuttings I am not sure if it's worth the effort, unless you're looking for long term storage or to sell them via the mail (there were some vendors that offered Salvia tissue cultures a year or two ago actually). That reminds me of another great use for tissue culture, it's way easier/safer to send plants in culture via mail, since they don't need light and are completely self contained.

It took me a lot of digging around to find good info on the subject, but its out there. Many people like to keep this stuff as a trade secret of sorts. Basically in vitro cultivation is a great last resort to save that rare dying cactus that is being consumed by rot, yet all that is left of the cactus is is some green skin tissue and one areole. Through in vitro, that tiny piece of flesh can grow begin to produce offshoots, soon becoming a whole new specimen. The methods aren't too complicated, but I've been unable to find a source for the required medium components.

This guy has a good assortment of videos that outline the basic principles, techniques and procedures that go along with microprop. It's really interesting stuff.
