agar said:

Pint jar on right is multispore inoculated (SA).
Allowed to colonize near 100%.
Syringe is 60 ML w/18 gauge needle.
Quart jar on left is sterilized water.

Aspirated 60 ML sterile water into syringe.

Injected 60 ML sterile water into colonized grain jar.

Shook grain jar to liberate mycelium from grain.

Injected total of 180 ML sterile water into grain jar.
(shaking mildly between each injection)

Aspirated out mycelium laden water from grain jar.
(all I could aspirate out of the grain jar was 150 ML)
Then, injected mycelium laden water back into sterile water jar.

Approximately 500 ML of Mycelium laden water.
(enough LC for approximately 50....10 ML syringes)

The advantage of this procedure is that you can be assured.
The culture in clean.
Because you can VISUALLY inspect colonization in the grain jar.
(if there is FUNK, you can see it )

That is not the case, with sugar type LC's.
(multispore syringe inoculated)
They can be contaminated, from day 1.
And, you NEVER know it. :frown:
Until you inoculate spawn jars with that LC.
Then, have every single spawn jar GO SOUTH. :mad2:

just so the newer guys, whom can't see the log section, can see it .

Should I make the injection port on the side of the lid when I make these?

I was thinking about making new lids to use for this tek, because it's hard as hell to aspirate liquid from a port that's in the center of the lid.

Do you guys think this would be a good idea if i tweaked my lids?

I've used this technique before, and I find that my colonization of new grains from a water based LC has taken a while to start growth whereas other sugar based LC's have taken 48-72 hours to see vigorous growth.

Any idea as to why? I have been using Creepers.

this is a great tek... I've been doing it for years.  you can use it to clone to (http://www.shroomery.org/forums/showflat.php/Cat/0/Number/6590986/an/0/page/2)

or you can use the same process to blindly reduce the number of substrains present in a MS (http://www.shroomery.org/forums/showflat.php/Cat/0/Number/5781218/an/0/page/3

you can also dry out those grain jars and store at room temp. to reanimate for later use... you need a filter controlling lid tho.. (http://www.shroomery.org/forums/showflat.php/Cat/0/Number/6886717/an/0/page/0)

nice extension of the tek, Agar.

Sterile water was first used to preserve cultures by Castellani in 1939 (2). Since then, many scientists have used this method; McGinnis et al. in 1974 (3) and Odds in 1991 (4) reported that they were able to maintain viable cultures for more than three years, without degredation.
