Psilocybe semilanceata - indoor cultivation * 2
    #4248496 - 06/02/05 01:32 PM (6 years, 11 months ago)

Psilocybe semilanceata - Indoor attempt.

Original wild specimen collected in Oct. 2004. I cloned this one because it was very white and unique.

Grass seed spawn of above clone was mixed with sterilized horse manure and placed in a microbox with filtered lid. A small amount of pasturized casing was added after full colonization.

Pins developing. It is hard to get a decent photo but you should be able to see them. Fruiting is at 58F and hyphal knots are everywhere, even in uncased areas. Sorry this post is so brief, I ran out of time. I'll beef it up early next week.

Very nice. I have only seen one picture of semi's growing indoors before. I might try cloning myself when they are up in a few months. I usually get them from a field with sheep so I suppose when I pick them I could collect some sheep manure too.

christ on a bike. i know if anyone can do it, you'll be the one - the only pic i ever saw was in Stamets of some libcaps in a conical flask. if it hadn't been Stamets, i'd have accused it of being a faked pic. i dunno why this species is reputedly so hard, i just always heard that it can't be done.

so you have it pinning from horse manure, and the only life forms present are the semi mycelium and whatever survived the casing pasteurisation. was that outdoor soil, or what? could there be any helper substances present? cos if not, this should be repeatable in any setup anywhere. i'm gonna have a go from a sporeprint i think, as i can't wait til september.

and i agree with the sheep shit comment, have thought the same thing myself.

Good to see, but pinning is a lot easier than actually primordia formation. After all, subaeruginosa, arcana and azurescens all pin inside jars at temperatures that i know would never, ever support fruiting.
Pinning can be a good sign ofcourse, but sometimes an isolate pins on agar, pins its arse off in the jar and then fucken vegetates out in the wood bed.

i once took inside a pice of "dirt" w some pins of what i thought was an inactive dung loving specie for studying its devolpment. But it showed out to be semils. It flushed second time 2-3 months later (in winter when my room was cold)

i took some pics of both flushes.

1st time in october some days after taking home:

second time in february:

I know this isnt growing.

Update 6/9/05:
Old pins in microbox are stalling out, but new bigger pins have erupted nearby.

Another modified attempt using the same culture. Grass seed spawn with sterilized grass straw (dried weeds from the yard) and a small amount of horse manure. A tiny bit of garden soil was placed on the cake after fully colonized to provide stimulating organisms.

Pins are forming only where dirt is present.

I've seen plenty of libertys that look like that when they're young. They have a more 'olive' kind of colour sometimes rather than that trademark psilocybe caramel colour of some other types. Although I've seen plenty of caramel libs too.

Those little suckers must be getting near maturity by now!

Not quite mature yet. The pins in the microbox that I thought had stalled are actually still growing. The caps seem to be a bit malformed on some of them.

Here is a little preview of my next post.

Psilocybe zapotecorum pins just emerging on cased grass seed. I expect them to be more impressive and photo-worthy by Tuesday.

Things are not going very well. The dark caps are aborts.

The problems may be strain related. I am going to use the knowledge learned with this partially successful experiment with a better (more normal) looking sample.

first of i would like to say thx for trying and publicating psi semilanceata
culture indoors, it was quite a thrill.

second i would like to comment a bit on your cake that seems as if it was
somehow 'inadequate' to perform proper fruiting.

after i have looked at your image closer, i found that most hyphal knots
were to be found on the dung (?) covered areas, it seems as if knot
formation would be easier on dung?

maybe try, if it happens to be dung, next time to try a dung mix (straw or something in addition)
or roll your colonised ball in some dung, thereby covering the outside,
incubate a day or two then fruit.

imho sure would be worth a try

I believe the darkn crap your seeing is actually a little peat sprinkled on...I think he mentioned they were pinning only where there was a little peat (put to get some micro activity going, I believe the post about it is above)

so it would be interesting to case it, not just in tiny areas, but as a whole.

What stroke me as amazing is the different phenotype in "captive cultivation" compared to the wild one!

Keep it up,you dont see Semilanceatas every day being cultivated succesfully!

I haven't noticed any sclerotia with this grow. I did observe some tiny stones form from an Austrian strain, but nothing impressive. I am aware of that Gartz image but it seems suspiciously staged to me. I have never seen mushrooms grow like that in a flask.

I suspect, and this is only speculation, that Gartz did grow semilanceata indoors but for some reason didn't take any pictures or lost the pictures of the experiment. His written description of his cultivation method seems factual. In an effort to make his publication look more complete, I think he may have placed wild specimens in a flask of colonized compost for the photo. Of course I might be completely wrong.
------------------------------------------
Features of the photo that raise my suspicion (I have a larger and more detailed version to inspect.)

No pins: There are only mature mushrooms with no signs of hyphal knots, immature pins, juvenile mushrooms or aborts.

Natural looking mushrooms: The mushrooms look identical to wild picked mushrooms. This is almost never the case with cultivated mushrooms unless they are under ideal conditions. A flask is hardly ideal and some distortion should be expected. Looking at other grows by Gartz of different species supports this.

Drying of lower mushrooms: The mushrooms on top are moist while deeper in the flask, where it is presumably more humid, the caps are showing the characteristic hygrophanous nature of P. semilanceata. You would expect the mushrooms nearest the flask opening to be experiencing the drying instead of the lower ones.

I thought it looked like somebody just dug up some soil and dumped it in the jar, but never thought of all the other reasons that as to why you doubted it too. I could create a picture which looks more authentic myself.

Second flush of microbox. The mushrooms are becoming more abundant but there are still problems with the form of the mature specimens.

Detail of separable gelatinous pellicle. This structure is a good identifying feature for Psilocybe semilanceata and P. pelliculosa. A separable pellicle is often noted in the descriptions other psilocybe species but is very dramatic in the two noted above. The pellicle in species such as P. cyanescens and P. azurescens is often difficult to remove. Tearing across the cap reveals a clear stretchy layer similar in texture to a soft contact lense. The pellicle is only visible with moist specimens.

Absolutely top work there.

Makes me all nostagic here in the UK we are fortunate to have semi's growing wild in the autumn.

I don't know whether its looking back with rose tinted spectacles but I'm sure the semi's I had as a 20 year old certainly seemed stronger than the cubes I occasionally come across nowadays.

Whoever can devise a simple tek for succesfully growing semi's like we can grow cubes should go straight to heaven IMHO so Workman you are almost there

First of all let me say, absoloutely fantastic. Great to see liberty caps grown indoors. I am about to attempt to cultivate them myself (even though there probably isn't any point, since they grow wild here; and by time my cultured ones fruit it will be the season anyway)

I have to say though, the caps you have grown resemble one end of the spectrum as far as liberty caps go; The small mushrooms always start out gellatinous and clear like you have shown - and indeed some grow large like that, but you are missing what I feel is an altogether healthier specimen; shown at hxxp://en.wikipedia.org/wiki/Image:Psilocybe_semilanceata.jpg

Your specimen is more like the ones on the right hand side; and indeed younger mushrooms look EXACTLY like the ones you have cultivated. I don't know whether this is a result of indoor cultivation, temperatures that are too high, or lack of temperature variation. I suspect the latter two!

For my own grow:
What temperatures were you incubating/refridgerating at? if not incubating, what temperatures for mycelial growth? and also, any further thoughts on optimal susbstrate composition?

Thankyou,

Thomas

The original culture for this grow was weird to begin with. It was very white and dome shaped when I collected it so it shouldn't be a surprise that it looks a bit off. The outdoor cultivated mushrooms were bluish when young.

I plan to do some work with some other more normal looking strains this summer. Incubation temperatures was normal room temperature 75F. Fruiting was around 55F.

Although 100% sterilized horse manure worked, it colonized extremely slowly and is obviously not ideal. A better substrate is grass seed with about 25% by volume moist horse manure. This colonizes much better and is similar to what Gartz used for his indoor fruitings. Unfortunately, this is not a suitable substrate for outdoors as mice completely destroyed last seasons test beds.

Good luck!

Great grow dude, I would definately be tempted to mix compost through the entire mix though. and case with unpasturized compost. What temperatures were you at for vegative growth?

Ah sorry you answered allready.

Yeah, I still suspect 75F to be a bit high in temperature. Even in the summer here we don't get much above 18*C and this is prime liberty terretory. However, I don't see why hotter wouldn't be faster. It certainly isn't reaching denaturing temperatures.

P.S could you take a look at http://www.shroomery.org/forums/showflat.php?Cat=0&Number=8239105&page=0&vc=#Post8239105

which shows one of my semilancaeta plates -
it has some strange ariel growing mycellium - notibly towards the edge of the plate too - which seems rhizomorphic and is colonizing at about the correct speed for psilocybe mycellium. I've just never seen it this vertically inclined! however, the plate has been stored upside down and the agar in that plate does lapse round onto the lid a little (my fault, too full.)

What do you think? Somekind of fungal contamination or genuine p. semilancaeta? there was one other plate with something simmelar to this, but imho not such a good specimen, and also very heavilly contaminated with a range of pink, orange, white and grey bacteria. Also, there seemed to be some other arial moulds, green mould and black mould.

Read the thread for the reasons why :P

That doesn't appear to be Psilocybe semilanceata mycelium and it is growing far too fast. It does look like some sort of mushroom mycelium but not like any P. semilanceata mycelium I have ever seen.

yeah, unfortunately it lacks fruiting ability.

I am going to order a p. semi print soon, from a reputable spore vendor;
along with some cyans, big laughing gym and some innocybe dealbata.

how do you know that it lacks fruiting ability when both you could easily have healthy P. semilanceata mycellia and still not get a culture to fruit for you, and the identity of the culture you do have is in question. if it is another organism and you don't know what it is, you can't know it's fruiting requirements can you? i suggest you play with it, a lot.
