[IN PROGRESS] Step By Step Cultivation Guide for Beginners BY a Beginner using Agar and Monotub!!

Hi all,

PLEASE NOTE:- I SHALL UPDATE THIS AS AND WHEN I DO STUFF.

Started this hobby for fun and thought of documenting the steps to make it easier for the beginners. This contains links to the teks used. Thanks to all the people who wrote all those teks.

Things Used :- (* is optional). You may choose to buy from anywhere you like or prefer. IN NO WAY I AM PROMOTING ANY VENDOR ETC. Provided links just to help people to identify what they are looking for.

1. 110 QT Sterlite - 1 Qty - Required for SAB
2. 66 QT Sterlite - 2 QTY - Required for Monotub
3. 4" PVC Closeout with Plug - 2 QTY - Required for Hand Holes and its cover in SAB. Sometimes you may buy these separately.
4. Silicone Tube - 1 QTY - To Seal Outer part of Hand holes and SAB.
5. Silicone II tube - 1 Qty- to seal the inner part of hand holes and SAB.
6. Tyvek Sleeves - 1 Pair - To cover the skinny end of the hand hole.
7. Tyvek Whole Coverall* - 1 Qty - To Cover Yourself while performing sterile work.
8. 3-9/16" to 4-1/2" Stainless steel clamps - 2 Qty - To hold Tyvek sleeves to hand holes.
9. 5 Gallons Buckets - 2 QTY - to Wash your Substrate etc
10. Fiberglass Insect Screen - 1 Small Roll - Can be used as Strainers for Multiple purposes. Used to Dry the Grain
11. Agar
12. Mini Glad Rounds. Put a hole in the lid using 1/4 inch drill bit.
13. Green Food Coloring. As suggested in PatsyWhyte's Agar Tek Guide.
14. Mash Potatoes Flakes
15. Karo
16. Micropore Tape :- This is to tape on the hole your drill in the mini glad round lid.
17. Aluminium Foil: To cover before PC. I hope you dont need  a link to this. :laugh:
18. PC
19. Gloves - Get it in any pharmacy
20. 70% Isopropyl alcohol - Get it in any pharmacy.
21. Butane Torch - Used to heat the syringe needle before inoculating.
22. Butane Fuel - To fill your butane light.
23. Light Malt Extract
24. Nutritional Yeast
25. Peptone
26. Ground Rye Grain - you can get this at local grocery or grind you substrate rye berries.
27. Petri Dishes
28. Scalpel # 11
29. Parafilm
30. Polycarbonate Erlenmeyer Flask
31. Organic Rye berries
32. Garden Gypsum
33. Wide Mouth Quart Mason Jars
34. Synthetic Filter Discs

WILL KEEP UPDATING AS AND WHEN I USE STUFF!!


1. Make Still Air Box. To Drill Holes I went to Home Depot and used their Drills on Display :smile:. The Home Depot Manager was kind enough to let me use their hole saw and arbor for free. They even helped me put the hole in the tote.



2. Make your Agar Mix for Inoculation. I personally used PatsyWhytes's Agar Tek.


3. Prepare Your SAB Workspace.

4a. Using this video to inoculate Agar with Spore Syringes. This is first time attempt. If this fails I shall use spore print. Wore tyvek coverall and gloves. Sprayed everything with Isopropyl Alcohol

Stored the mini glads in closet after inoculation.

After 3 weeks of inoculation, I took some pics. Here it is. Dont Know what the yellow thing on the bottom of the container is. Need Help here:-



Agar to Agar Transfer :-

In the Pics above, the agar transferred from Green to Yellow. This is just Mycelium waste and is perfectly normal when you add color to your agar. It is not Bacteria growth. The agar is loosing nutrients and eating the color. Its time to do a transfer. (Thanks for all the experts- Numero, SpaceHildo, Grey and others) for guiding me here.

On 06/05/2015 I did Agar to Agar Transfer. This time I used MEYPA as per the recipe from Paul Stamets Book:-

For 1 Liter of water :-
20 gm Light Malt Extract
2 Gm Yeast
5 gm Ground rye grain
5 gm Peptone
20 gm Agar

Video Link to make your Agar Mix.

Mix ingredients and put the mixed contents in erlenmeyer Flask. Then Add water. Screw the top with a vent cap. Then PC at 15 psi for 30 mins. Once sterilized let the PC come back to 1 Psi naturally. After that transfer the flask to your sab along with petri dishes. Let it cool down and while the agar is still liquid pour your petri dishes. Once the Agar solidify make the transfer using the video Agar to Agar Transfer

I made enough only 250 ml of solution and poured the petri dishes. Once agar to agar transfer was made in SAB, I sealed the Petri dishes using Parafilm. Some people also use saran wrap instead of parafilm.

As of 06/14/2015 the growth looks as below.

Finally I decided to use the following plates and sectors for Grain Inoculation:-

I Used the Rye berries as grains here. use the following step by step guide for Rye Berry Preparation

It says SWIM, then says "things I use".

I don't see too many working display drills at my hardware stores. I can't imagine they would be cool with you drilling that tote.

Haha oh grey. Would be funny to see a guy huddled in a corner at home hardware drilling holes into his totes. I'd be like I know where you're doing :wink:. On a side note my home Depot has a tool rental area. Could just rent a drill and hole saws for a day for pretty cheap

I have the following questions guys.. Any help would be appreciated...:smile:

1. If you see the pics, the mycelium on top itself is pure white. However when i see the mini glad from below it has turned yellow. I am not sure if it is contaminant or some mycelium excreta?

2. if the below is contaminated can i still use the mycelium from the top?

3. If yellow is contaminant, i still have some portion that is still green. Can i salvage that and use that for further process?

4. The Next process from any part if salvaged.. Will it be creating more agar plates or inoculating the grain jar?

I got some replies in PM. Some said its normal and some said its bacterial contamination.. So was confused..

Anyways... what do experts suggest I do now? Is my agar ready for grain inoculation or should i do another transfer and then do grain inoculation?

people who have to pm their replies probably knows their advice would be shot down
by people with actual experience. I'd take all replies coming in pm's with an even larger grain of salt than replies given on the boards.

you can do whatever you want with the agar plates. the pics of the myc is way out of focus for me to tell whether they're clean or not
but its your tek so you do whatever you want your tek to show man! :super:

Its not about show.. I want to do it right..

Is it safe to open these plates in open air to take more focussed pics..
Also all the learning from these queries will be consolidated in the tek.. so that people who follow this in future are not confused.

I can post more clear pics..if i can open the glad rounds.. the material of the glad round is translucent and hence the cam is not able to focus..

If the color is getting eaten up, those plates need to be used/transferred before the mycelium runs out of nutrients.

Don't open them in open air. You can open them in a SAB. If you have headphones for your phone that has the little microphone w/ button on it you can use that button to take a picture. Just turn on your phone camera, set the phone on the top of the SAB looking in, with both hands in the SAB open the plate, and press the little button with your mouth or some other creative way. I've done it before, but now my SAB is all scratched up so it's hard to focus.

Before i inoculate grain jars, i just want to be sure if the myc is correct.
@Grey: Looking at the current pics, can you advise if this is good enough?

Else Once i am done work i will go and open it in sab and post some pics. Based on the advise I get, i would either inoculate the grain or do a transfer.

I can't say I haven't used a plate like that, but I prefer to have clean/less random looking growth. Make a transfer to be safe, but you could still use them afterwards if your comfortable with your sterile technique.

1. What are the health growth Sections as per you?
2. How many plates do you suggest to transfer to?
3. Do i discard the rest of the plates or let them grow?

Avoid anywhere that is obviously bacterial. Some have clean growth, but There a couple nasty ones I'd avoid. The first and last plate both have bacteria, but also have good looking growth. 12 o clock on the first plate and nine on the last. The second and fifth I wouldn't bother with.

If it has bacteria, don't let it grow. Trash it. Your in the process of getting a clean culture. Once that's a success you can expand further.

Where is bacteria in the first plate? I wish to know, how to identify bacteria.. I thought bacteria would be yellow. I will discard the 2nd and the 5th plate. What about between 4 and 9 in the third plate?

6 o clock on that first plate. The mycelium is very weak there. Even if it were clean, I wouldn't transfer from there when directly above it looks great.

The third plate has good growth, but also has weak or spotty growth. 6 and 8-8:30 on the plate is where the mycelium fans out looks the best to me.

First has those gross bacterial spots around the edges of the plate from 12 o'clock the 3. I wouldn't use this one since you have clean plates.

2nd has a big bacterial blotch at the 3-6 o'clock position

3rd looks great for grain inoculation and transfers from 1 o'clock to 7 o'clock.

4th looks good for grain and transfers from 10-5 o'clock position

5th is too disorganized for my liking

6th is clean and healthy and looks aggressive everywhere except the right hand side in my opinion.

I've never built an engine. I've also never written a guide on how to build an engine. If I were to write a guide to build an engine, it wouldn't be as I was building my first engine.

If I were to build an engine for the firs time, I wouldn't want the guide I was following to be written by someone who has never built and engine, or who was in the process of building one for the first time.

You have to see what I'm getting at.

its the plates you put the wedges you cut out onto that's interesting for us.
those plates there shouldn't be used. and you should have taken smaller wedges, as small as you can.
