Agar to LC test tube!!! Thoughts 1

So I was bored today and decided to come up with something to help me go from agar to LC so this is what I made!!
Silicone injection port at the top then a vent hole on the side of the tube with a tyvek patch taped on with micro-pore..... I was thinking put some agar in it then PC, use an inoculation loop to transfer to the test tube from agar in Petri, then inject honey/sugar in the raw/malt extract or whatever with a syringe... Give it a shake then draw back into a syringe....
Thoughts?!

when you're done pouring plates leave a little agar in your bottle. drop a wedge in let it colonize. squirt water into your bottle shake around and pour into jars.

Hmmm.... I've havent done any agar work yet. I have 10 pre poured PDA dishes and 40 Petri dishes with 4oz of MEA powder...
I'm gathering all my supplies now... I think I have EVERYTHING but a flow hood.

you don't need a flow hood unless you're operating a small sized farm. if you're not doing 100s of plates a week and making more than a few dozen jars then it's pretty useless at least to me.
I don't have one because I don't need one and I do plenty of jars and agar work.

You probably already know about it but grain LC is super cool. Colonize a jar of wbs, inject sterile water, shake, draw mycelium rich water back into syringe, transfer. Cuts out the agar work completely.

I would still do the agar work.
I made a blank water syringe with a few drops of honey in the pot. Squirt it into the agar plate, drew back up, and shook daily. There's a bunch of mycelium in there now. I haven't tested it on agar though, just a couple invitro test cakes. I'm sure it could work the way your thinking.

Never knew about this... I have 2 grain jars 25% colonized now and a syringe of sterile water!

Agreed! Hope all goes well... I know the AA+ is legit. The atl seemed very light..
Some proof and not sure why the random gold cap but everything else was on..

Planning on making 40qts spawn. If these fail I was gonna fall back on the PESH. If that fails, I'll just do another b+ grow.

I have about 5 vendor syringes on deck if your in need... since your not far from me. KSSS, Creeper, blue meanie, Treasure coast, pink buffalo. I'm using them for my agar in attempts to cut down on contams from the beginning.

I do like this idea. I was dropping agar wedges to brf cakes the other day and something similar was sugguested, to squirt some sterile water on a plate and agitate the myc with a needle and then suck it back up.
I like using 1/4 pint masons for agar so I think itd be easy to just pour into a few jars, drop a little wedge onto those
Kind of a quick liquid innoculant for test cakes

Yeah Munchausen had a video showing "easy li without a blender". If you squirt it hard enough and make a motion with the stream you can sort of pressure wash mycelium off the agar.

Flowhoods are useful and you'll definitely enjoy having one but don't let the lack of one slow you down. I do most my transfers at home in open air and have good success...found after a lot of SAB work that aseptic technique was enough. I probably lose a few more jars but I believe I make better time overall so it's worth it.

A flowhood and a pressure cooker are 2 major tools that make life MUCH easier in this hobby.
Working with large spawn bags inside of an SAB is a highly awkward, and a virtually impossible task to perform.

Doing this in the past seemed impossible to me...

Ive been looking into small flow hoods and glove boxes and thing a hood would be a better choice. something like a 12x12 would be enough for me considering I'm just a hobbyist. I've seen a few portable flow hoods and mini clean rooms that would fit my needs nicely though.

Go for 24x 12,a 12x12 will really only fit agar work comfortably which is fine but the real convience comes from g2g jars and spawn bags, every bit of work can be done easy with a 24x12 and its still small enough to move around with you. My 18x24 has handles but its a bit bulky I dont like to move it around too much I just use my still air box in those situations
You wanna keep your lab area clean and you do wanna use upmost sterile procedure but I dont think you need a full clean room just reasonable precautions. The space right in front of your flow hood is your clean space. Run the hood for a little while to scrub up the air some
I know RR has stated that for years he ran his flow hood in a bedroom with carpeting, that his dog slept in
