Learning Objectives: -Establish a novice practical knowledge of WBS jars (preparation, inoculation, colonization, & spawning. -Establish a novice practical knowledge of coco coir bulk substrate (inoculation, colonization, and fruiting). -Gain an understanding of the technology and science of the Monotub. -Successfully construct and utilize a Monotub -Provide exemplary information on bulk cultivation for new cultivators and curious minds alike.
Materials Needed: -Pressure cooker -aluminum foil -MS Syringe containing spores from unnamed species/strain. -quart mason jars with rings (5 8 for this first attempt) -prepared WBS lids with SHIP (self healing inoculation port- x2 -jar lid -drill with 1/4" drill bit -tyvek (in this case a tyvek coverall $3.50 from Wal-Mart) -High temp Silicon gasket maker. -polyfill -medical micropore tape -prepared WBS lid (tyvek filter and polyfill stuff inoculation port) -x6 -prepared wild bird seed (WBS) -coco coir (exact amounts coming soon) -vermiculite (bucket TEK show course Verm) -constructed monotub -polyfill -sharp knife or shears -impermeable tape -can liner -SAB (still air box) -sanitizing agents (isopropyl alcohol, bleach spray, paper towels, alcohol lamp) Please let me know if I missed anything.
Welcome back to another mycologist217 grow log. This time we will be cultivating an unnamed species of mushroom using Doc34's Wild Bird Seed TEK , and The Ohmatic Monotub TEK along with his coir pasteurization TEK. however, I will be using This coir substrate prep TEK (I will still be using "the bucket TEK" for pasteurization); Franks Simple Coir/Verm TEK for substrate prep, and Franks spawning TEK
I've begun the grow by starting the soaking of my WBS, and constructing my jar lids. More updates coming after inoculations.
I've decided that only one two of the jars will use a lid equipped with a SHIP. I've used this design (with very successful results) for liquid cultures before and will use it for one of my WBS jars. The other four jars will use the procedure outlined in Doc34's WBS TEK. Here is a picture of my ship design:
The metal tube is a brass plumbing insert that has been gasketeed to the lid with high temp silicon. The bottom has been crimped and the insert has been stuffed with a wad of polyfilly before having a square of tyvek taped over it with micropore medical tape. The red spots visible are the SHIP (self healing injection port) and are simply high temp silicon that has been neatly squeezed through the drilled hole.

Day 0-Day -1 - This afternoon I finished preparing my WBS by draining it, rinsing it thoroughly, simmering it, then letting it drain dry for some time (this trial was roughly 55 minutes). It passed the toilet paper test.
I loaded my quart jars roughly halfway with the prepared WBS before applying the prepared WBS lids (in this case 2 with gas exchange port and a SHIP, and 6 with a polyfill filtered monoport over a tyvek filter). The jars were pressure cooked at 15psi for 60 minutes. After pressure naturally normalized, the jars were shaken and left to cool.
Inoculations are to take place in roughly 18 hours. For now enjoy this picture of my prepared jars presterilization:

I wouldn't use up all your spores because standard practice says sterilize for 90 minutes, not 60.
I did 60 for a long time, not to say it won't work(sometimes), just maybe don't use everything up

Day 0- I inoculated my wild birdseed jars this morning. I must confess I'm very nervous about this whole procedure, but only because it's my first trial and I'm not familiar at all with what looks good and bad and where corners can be curved or where there is room for error. In my overall cultivation experience until now, I've found the mycelium to be forgiving and resilient.
it should be noted that when removing the needle from one of the 8 jars, the polyfill filter was pulled out of the lid. I micropore taped a piece of ISOlropyl soaked polyfill over the monoport
I did inoculate with a MS syringe. Can someone indicate to me how long it should take for mycelial growth to become evident?

Day 3- I'm pretty sure I can see mycelium growing in at least one of my eight jars. Honestly I wouldn't be surprised if I messed this all up. I feel like too many of my kernels are exploded...and I have no idea what I'm supposed to be looking for. Only time will tell. At least I have my healthy PF TEK grow coming along to keep my mind at ease.
I picked up some gypsum and perlite today to restock, and I also went ahead and bought a new bag of WBS from the local Kroger...a huge value...only $4.50
If I am forced to start over and inoculate new jars, I will construct SHIP bearing lids for all my jars as currently the only jar I'm pretty sure has visible mycelium is one of the two jars with a SHIP.

I have issues with WBS aswell, and identifying issues in general, day 8 now after inoculating WBS with creeper strain from spores 101.

Day 4- Please to discover tonight that 7 of my 8 jars are sporting patches of new mycelium. Still think my seeds might be too high in concentration of exploded seeds...don't know...but I love these jars and are sending them all the love I can.

Day 6- Upon inspection today all 8 of my WBS jars have mycelium growing in them. No signs of anything but white currently! I gave each jar a decent shake today as directed by the TEK. I must note that I really enjoy observing this colonization more than the PF TEK jars. It might be the interactive nature of giving the WBS a good shake ever 4-5 days or so.
I have started prepping more WBS for another 8 jar culture run. Really want to get it right the first time and I feel like maximizing my opportunity is the best bet. No new pics as of yet as there is little to get on camera yet...much less than meets the eye in person. More pics she the colonization reaches more notable points.

Day 6- The mycelium in many of the jars was effectively spread across the jar during the shake. There are now patches throughout the entirety of all the jars.
-it should be noted that the jars have been kept at 80D F pretty consistently.

Day 7 - My jars are simply exploding with growth, and as the growth becomes more and more expansive the rhizomorphic bits of the mycelium are becoming more and more apparent. It's really exciting to watch. I would estimate my jars at around 50-60% colonized but I'm not sure. At this rate however, Ill be ready to spawn them on Easter or early next week. Never imagined I'd see this type of progress on my first WBS inoculation but I understand the real test is during substrate colonization here is a picture of one of the jars:
Also I finished prepping my next quarts of WBS jars this morning and have them pressure sterilizing right now. I did build eight brand new grain/WBS lids featuring SHIPs, the design of which was posted earlier in this log. But here they are before going into the sterilizer:
Of course I added aluminum foil lids before putting them in PC.

Looking great!! Also on a side note bulk is actually way easier and a lot more forgiving. There's a reason why all you need is a pasteurization rather than sterilizing like with grains. It's because grains can host LOTS of molds very quickly. Out on the fields of manure, you won't see much molds on the old aged manure and coir is basically so contamination resistant you don't even need to pasteurize properly like with the bucket tek
In my experience bulk grains have always been the biggest challenge

Day 8- The jars are colonizing at what I consider an unreal pace. I feel like they will be at 100% colonization by Saturday, a coup,e of the jars seem closer to 70-80% colonized already. I gave each jar a good shake today, and I suppose now I should just what until 100%? Obviously I want to spawn my jars together as part of the same Monotub. If my jars colonize a couple days apart is it going to be okay to let the faster jars wait?
What is the average colonization time for a WBS jar inoculated from MS syringe? Is 9-10 days average? Heres a picture of one of the slower jars:

but i would like to point out those grains look a lil too wet, over cooked, and starchy.

Starchy? All my passed the tissue test. I feel really good about these jars. On my way to a bountiful first Monotub. I feel it in the air
