Nice videos man. I'm happy to know that I do most of the stuff you're doing in there the same way. I don't put my plate upside down though, although I do open them upside down in my hands when looking for stuff.

A few questions:

1. I find that having the spore syringe so high when letting the droplet go often creates a splash in there. It makes it look like I have satellites or promotes weird growth. Do you get this problem?

2. Why don't you cool your blade off in the receiving agar plate? Wouldn't the hot blade damage the mycelium you're cutting into or is that just some overly careful BS people spew?

nope

2.I use an x-acto blade. Its super thin and cools off pretty quick. I'd never bothered to try to cool it, and never had any problems. I'd imagine the fixed blade scalpel you see in RR's vids has a bit more body to it so it might cool down slower than my little disposable blade.

Hi Munch,

I actually don't think you can eyeball the splashes I'm talking about. They're very tiny and show up when the mycelium starts developing. But I guess you would've noticed this beforehand as I'm sure you've done many single drops from that high up.

As far as blade cooling, I also use an X-acto blade and it takes me 3 dips in the receiving plate before it stops making the "tssss" burning sound. But I'm sure it's not that big of a deal.

in the agar to agar video it looks like you may have put your left hand on the receiving dishes' lid, hard to tell from the angle but it may be behind the lid too. hands never above sterile surfaces.

I've only ever had this happen when I get too much on the plate, like more than one drop, which is easy to do. Not all my syringes flow as smoothly as the APE used in the video. But when I do the single drop, they all germ nicely in the middle. Is it possible you might have tilted the plate and shifted the water?

I just heated my #2 xacto blade and it was cool to the touch after a 20 count. Here is a video showing that my blade has cooled to the touch by the time it hits the agar in my video.

Yep, like that, and I'm sure it's possible I tilted it. I'll do some more with single drops soon. I also use my X-acto immediately after it's flame sterilized as I thought it was supposed to be used ASAP after red hot.

oh shit you are right. good eyes. I probably didn't notice it when it happened because I was watching myself on the video screen to make sure I was in frame, rather than concentrate on my hand placement. I will have the video replaced later today.

I usually do too, but just the process of turning my torch off, putting my hands in the sab, and then grabbing the plate all takes roughly 15-20 seconds, so its cooled down by then. I'm not doing any active waiting for it to cool down. RR drags it through agar because he has a much more convenient setup with his flowhood, and he doesn't have any time between flaming and working like I do with the SAB.

just for safety reasons and because I can do multiple plates with the syringe since there is no contact. I get scared that I might knock it off my table and then it catches my house on fire. I get cases of power 5x for $20, so its def not me being cheap. PF-tek, I leave it on and do 5 jars at a time, turn the torch off and take a breather and do a dab or something. I fucking hate pf jars.

I work in a SAB and have a standard alcohol flame and it takes me like 2 seconds to move the blade from flame to agar plate.

can't be too careful :shrug:

clean long sleeves and tape, next best thing to tyvek sleeves when I'm out. I figured most people probably don't have them, though.

Ah okay.  I kinda hate the tyvek sleeves sometimes when I use them.  I've taken to just using my elbow length gloves and trying not to get my whole arm in there most of the time unless it's super serious.
