4% is a little stiff for spore germination IMO. Usually I only do pour agar for antibiotic and I save that for wild prints and clones at which point I usually aim for around 2.5%-3%. Plus its so dry here that if I make it too stiff it cracks and dries out super fast. Climates less shitty than mine might do fine with a full 4% agar.
I also find that the media used will affect it. For example IME cornstarch will tend to make a stiffer agar while oatmeal agar tends to be softer. Experiment to find what combo of what works best for your individual situation

I haven't had any issues thus far with it.. if I do I'll try lightening it up a bit. I am doing no pour though so I usually have some moisture from condensation and I usually pour them deeper than I would a petri.

blindingleaf said: pasty how much agar do u use if u are making a sleeve (25) of regular petris? for every 500ml of water i mean.
Are you using MEA powder or making the medium from scratch? If MEA powder then 20 grams per 500ml is just fine. Never mixed it all from scratch myself though. I did 8 plates tonight with 7 grams of MEA powder and 180 ML of water and it was just fine. You can fudge it a little.
i have done all pre made MEA up till a week ago. i loved it, so easy and perfect every time. just wanted to venture out so i ordered agar powder and LME. i made dog food agar, but used double amt of agar i should have. 10g agar power to just ten petris! they were stiff.

this is what i will try with some rye grain soak water too. i was adding half grain soak water to my pre made MEA and it was working really well. it was the only way i could think of to change the media.

How do these plates look to you guys? This is after the first transfer. I have had a problem with condensation as you can see in the pictures.
The growth looks rizomorphic on the flat part but it also looks like it is pretty fluffy. Do you think this is just ariel mycelum or is this possibly cobweb? Is my agar to rich in nutrients?
I know its probably hard to see the growth well from the pictures because of the way the glad mini rounds are not very clear, but it was the best I could get.
The slice I transfered to this dish stuck to the side without me realizing making it easier to see the rizo growth, but the other side is fluffy like all the others.
I want to start a few master jars, do you think that it will be ok to use some of these to start those (to have for g2g) and transfer a few further on dishes for an additional grow?
Thanks Pasty, Frank, and everyone. Very helpful teks/information!

yep. start some masters keep your g2gs and cultures labeled, fruit them out and see what you like best. it will still be semi random until you get into some isolates

Yah. this is a great tek. It makes agar seam much more accessible to new growers. I just started pouring pre sterilised petri dishes but I still plan on using pasty plates to germinate spores or take clone tissue before transferring to real petris.

this is EXACTLY what i use it for!! also been using it to get pins for cloning. I'm noticing that if i make my agar real stiff, like 20g to 500ml water instead of 10g, the culture pins pretty easily on the dish at around 4 weeks. i never had that happen on regular petris, no matter what the agar recipe

i just used this tek right after reading it to make amaranth soy agar. from the time i left my house to the time i finished making the lids it took me an hour and a half, and 40 mins of that was driving to and from walmart and sitting in line. now i have 8 rounds of agar that i'm ready to pc as soon as the silicone i used to seal the tyvek dries. thanks again!

silicone to seal the tyvek to the lid for gas exchange instead of micropore tape

I scratched up the plastic on mine so silicone would stick, and stuck SFDs on there, I don't wanna fuck around with chances.
