I need your help regarding using rye

Hello,

where to start .... well Iam using clones from a petri dish and not spore syringes. First the rye is colonizing pretty well and all white but always after around a week I can see green here and there.

The weird thing for me is that the green doesnt seem to appear somewhere in the glass but always also where the mycel is growing. Its not much but you still can see a little green / dark green.

This really gets frustrating already, first always everything seems fine and then green appears after a week and all work is for the trashcan. So my guess now its a problem because of endospores ??

How to fight them ? Maybe I really should let my rye soak up 24 hours ? Thats the other thing, I always read different things in the net and I also asked it once in a different forum and they told me its BS to soak it for 24 hours.

But then Iam reading somehwere else its needed because of the endospores. What Ive tried so far was always with a few hours soaking up to 12 hours with mixed results but I always had to throw away probably around half of the glasses or more.

So Iam asking now here cause I think most experiences users are in this forum, and btw sorry for my English.

Maybe I also tell you quick how I prepare the rye ...

I let it soak for a few hours, mostly around 4-6 but I also tried 12 then cook it for around 30-45 minutes. Then i let it sit in a collander and stir it now and then till its totally cooled down. Mostly thats around 2 hours till I cant see any steam anymore.

Now I just put it my 440ml glasses (polyfiled lids) and cover them with tin foil and put them into my pressure cooker for about 90 minutes. Now I let it cool down and use it after around 6 hours.

I know its always hard to do a remote diagnosis but still maybe you have a tip or two :smile: that would be really nice. With BRF Cakes I dont have any problems only if my spore syringe wasnt well made but I really want that rye also works because of using clones and so.

Let it soak for 24hrs, then pressure cook. See if that clears it up.

how are you inoculate'n your grains.....your prep could go like this....20-24 hr soak..... 5 min or less quick boil /just get the grains hot....strain..spread out on towel close to 1/4 -1/2 with fan...or if it cold out side ...that works,.... should dry right out ...load jars and pc 90.......be sure to shake the extra moisture around after they cool

Your cultures are contaminated.

Soaking is to hydrate the grains and to 'help' germinating endospores, which are bacteria and not green mold.

Since you're getting green mold, it's your culture that's the problem.

Hmm probably you are right, then my "technique" to transfer the clones from agar to the grain somewhere must be to "dirty / unclean". Iam using right now a glovebox but without gloves so the box is with 2 open holes.

I think Ill try to build something which will be totally closed with gloves on it.

Thx for your answer so now I know I probably dont need to mess with my preparation of the rye. And for the endospores part, thx for clearing this up to me, I always thought this also will cause mold and so, so its just for bacteria. I rarely ever have problems with bacteria, its "just" the mold which drives me nuts.

Probably not the rye. Just my opinion. What gives it away is the green is only where you shot it. Next time leave a small jar of grain un injected and you will most likely see nothing grow till you inject the green into it.

Iam happy to say only the first glasses which were around 8 were contaminated. This sounds alot but its around 15% of all glasses Ive made, the "funny" part is all the first glasses were failures and all others are till now just white xD. So I first thought all are contaminated again like always when Ive tried with rye. Sure it still could be better but Iam happy for now.

Iam sure it also had to do something with my knife I was using for the clone transfer from agar, it was just a normal thick knife. I really never could flame it till it was going hot red cause it was too thick. Also it was hard to work with it with the agar, now I have a real skalpel which is so much better.

Anyways I have a different question now so I will hijack my own thread instead of creating a new one if thats ok ...

I want to let the substrate later grow on straw. Ive seen the techniques with the plastic bag and the basket. I have now a question, can I just add the straw and layers of colonized rye to a plastic box and let it grow there ? Wouldnt it be just better ? And if I can do it, do I have to let  the lid open from the box while the rye is colonizing the straw ?

And do I have to mist the straw a bit from time to time while its colonizing or just leave it till its fully colonized ?

When Iam doing the plastic bag or basket technique, dont they need a high humidity enviroment ? Or can I just let it stay open in my room ?

I hope you understood what I was asking for, thx in advance for all help.

You can use most any container for straw, but be sure to place a weight on top after spawning to keep it compressed tightly during colonization.

Alright, I guess Iam trying  then different things the first time and see what works best for me.

Just a last question, when the rye is colonizing the straw do I also need to fan and / or mist them daily ? For BRF cakes I have the polyfiled lids so my guess is yes they also need fresh air with holes or faning right ? What about misting ?

And maybe a real last question :smile:, how much spawn in weight approx. do I have to use for how much weight of straw ?

Thats it for now, maybe I will post pics if I can lend me from someone a camera if my project has worked in this thread.

Hmm I really have an important question about the straw Ive bought. I could answer me the others already by reading alot in the last days.

Ive bought straw from Amazon here in Germany, it is barley straw usually bought for rodents. Now my question is, not everything in there is yellow like Ive seen in the pictures and it keeps me a bit worried now. There is a bit green here and there, should I be worried ? Or is it just normal ?

I firstly thought that there may be hay in it, but it isnt its just a bit green here and there.

That just means the straw isn't aged enough.  If possible, spread it out in the sun for a few days before shredding for use.

Damn I hope someone can answer me quick, I pasteurized the straw (what a mess btw :P) and now what ? I mean the straw is totally wet, should I let it sit a few hours or maybe a day till its a bit drier or use it now immediately ?

Cant find an answer to it,

EDIT: Well after over 4 hours it still was kinda wet, I let it drain and there wasnt coming anymore any water but still the straw itself was pretty wet IMO. Ive used it now anyways, hopefully it wont fail now but Iam really worried :frown:.

Hello,

everything went fine so far and I can see already some small mushrooms on top of the straw :smile:.

Now I have 2 questions if you don“t mind, how will I remoisten the basket case after the flush ? Just mist it ? Or let it flood under a water tap and then toss out the excess water from the bottom ?

Other question regarding harvesting, do I have to harvest every single small mushroom ? Or just the small aborts with the ones that are done ?

You can dunk them in the bath for 24 hours between harvests to rehydrate your substrate. I put the whole tub in there with a weight to keep the sub submerged. Now this obviously depends on the size of your container

Ah ok thanks for your answer. The baskets Iam using are tiny ones, I could put it into a bigger container, its no problem.

And what about harvesting, I did so many grows with the PF tek and never really knew how to handle it. Do I have to harvest only the aborts + the ones that are done or any mushroom I can still find on the straw or whatever Iam using at the moment ?

And again mold :/. Always when bulking or casing everything grows white until I put the casings or like this time the baskets into fruiting mode. After 2 weeks there are coming small mushrooms but also mold with it, but why ?

I really dont understand the reason behind this, maybe the colonization time with 10 days is too low and there is still substrate which isnt colonized and this gets contaminated ?

I really have no clue anymore, this is now really getting on my nerves :/. Or was my straw maybe to wet ?

If someone had a few tips or had some clue why this happen I would be very thankful.

10 days is good if things are right...you don't need caseing...but if you did...what is it and how did you prep it

Ah sorry for my bad wording. I didnt case the straw baskets in any way. I was just saying that it doesnt matter what Iam doing, if caseing or the straw tek both have the same result in the end and I really dont know anymore why.

What happens is this, I let the basket colonize in a plastic box aka terrarium and the plastic box itself is surrounded by a garbage bag so its in total darkness (same when Ive tried with the coco tek but then obviously not in baskets).

The plastic box has alot of holes drilled in which are filled with polyfill for gas / air exchange.

Now after 10 days everything looks white and pretty good, now I take away the garbage bag and fan the terrarium few times a day.

After 2 weeks I see some small mushrooms but also always at one place mold :/. Now you will see the mold growing rapidly :/.

This until now happened all the time for me, this time with straw and last times when Ive tried with the coco tek.

The straw tek I was using was the one from the RR DVD, with baskets.

Now a few questions coming to my mind ...

Maybe I should let the drilled holes open instead of polyfill them ? Iam really not sure about it.
Maybe I shouldnt use perlite for extra humidity in the plastic box when using caseings or for example the baskets ? Maybe its just too much and the caseings or straw getting too wet from it ?

When I threw away the straw from the baskets it was damn wet and warm in the middle of it.

What could be the problem ? Or just a simple problem of preparing the coco or straw ?

As far as I could see the straw itself didnt contaminate but in the middle of it the rye was. But I really check it twice when I open up glasses with colonized rye and all were not contaminated when spawned into the straw or coco.

Any help or tips would be really appreciated and sorry for my poor English sometimes :smile:.

how did you inoculate your rye? spores or  LC? yes ,if the straw was to wet or coir ,you may invite contams...if it is a sgfc,and if you got perlite =no polyfil

I cloned a mushroom on agar and put then the grown mushroom from the agar to my jars. After they were fully colonized I did some G2G transfers.

I still have some casings, baskets and monotubes going, they are now in fruiting mode and look all fine. Lets hope these are working, this time I removed the polyfill from the SGFC.

How about monotubes ? I also have some monotubes going, I think here I need the polyfill for the drilled holes or ? Cause without they would be too low on humidity or not ?

With PF-Tek, G2G, agar and so on I really dont have many problems anymore its always just the last phase of growing on bulks or casings.

I will see now if the new monotube and straw project is working, I did here much better with the preparation. The first time I had some problems with the temperature of the water.

But then again Iam wondering why everything looks always white and then if the first mushrooms are appearing you also see the mold coming :/.

Really thanks for your input, every tip/hint is welcome.

So after ALOT of trying Iam 100% sure that the mold is always coming because it was always too wet. When there was mold and I wanted to throw it out and I grabbed in it felt totally muddy at this spot.

I have a kinda montub right now going on with coir and it works perfect without any contamn and alot of shrooms. This time I took alot less water and took out the polyfil out of all drilled holes.

But now Iam wondering what to do with the small baskets, these sit now in terrariums without any perlite and no polyfiled holes.

Iam now scary to moisten at all lol, Iam now not sure if the water from the baskets will be enough to moisten the air ? The baskets fill around 60% of the volume in the height and around  80% of the width of the terrariums.

I just know now when I polyfil all holes and put in perlite I will get always mold after a few weeks. The question is now, will this also happen with perlite + opened up holes with no polyfill ?

Iam not sure why nobody could help me so far sorting this out. Mind you this isnt the first forum where Ive asked for help and nobody ever mentioned anything about moist lvl.

Ive come to the conclusion moist lvl matters even more than cleaness (is that even a word ?) in this phase of growing.

Ive made a Pic from my terrariums with the baskets in it, maybe you will get a better idea then what Iam asking about. Bear in mind Ive made this pic with a real crappy webcam, Iam sorry for it but I dont have anything like a digicam or something :frown:.



Like Ive said crappy quality but its just to show you the terrarium where the baskets are sitting in. You also can see the drilled holes in it, at least from the bottom tube. The top tube has the same but you cant see it because of the stupid lighting :frown:.

Now I dont know if I should put perlite in it or not.

I just know everytime I put perlite in AND stuff polyfill into the holes the whole thing contaminates always after around 2-3 weeks.

Right now everything is white but Iam worried about the RH.

You cant see any waterdrops on the boxes but the windows are always a bit steamed or clouded up, hopefully you know what I mean.

Nobody can answer me if I should put the baskets in a SGFC (made a SFGC out of the boxes from the pics now) with perlite or no perlite ? I really can“t find an answer to it :/.

I would put a basket like that into a SGFC.  Use something to elevate the basket about 1" above the perlite.  Also make sure that your SGFC has holes on all six sides and is elevated about 2" off of a desk or counter top.
