hey everyone just wanted to say this thread is kicking ass!
loving the shots from everyone, this makes me want microscopy to have it's own thread, maybe encourage more people to post useful details and DIY teks for microscopy
besides that..
so heres some shots i took the other night
a cross section of coprinus comatus
closer..
this is a pine stem cross section, not mycology but how bout that dark field eh?

I have a camera -> microscope adapter otw... It has a 2x lens, and with my 18mp Canon DSLR, that should = to atleast 15x  Ill be REALLY active here in a few weeks with pictures!

Given that the insides of the spores are moving around, I am guessing that you didn't mount that in pure water.

I can't see videos on my slow connection, but the inside of viable spores are alive and moving around rapidly. This movement really picks up in the minutes before hyphae emerges from the spore. I have a few videos showing this. You also see movement within the individual mycelium cells too. RR

I always thought that was due to brownian motion. A few days ago I looked at growing hyphae with GFP tagged cell wall synthesis enzymes and I could see vectorial movement of tiny round vesicles (I guess membrane vesicles carrying the fluorescent enzymes). Has anyone seen that in brightfield?

WOW!! AMAZING!!!
I didn't look for the movement until I read the comments. I wondered why you posted a movie of spores just sitting there. 
That pale thing in the center must be the haploid nucleus. Are those dark things moving around proteins that are eventually going to migrate to the cell membrane to shoot out hyphae?
