No-Pour Agar Agar TEK 13

I know a lot of you guys might not have a flowhood so I figured I would let the secret out on how I used to skip the pouring step when making agar plates.
Working in a glove box can be a pain and the less you expose your cultures to the still air in them the better. ------
Materials needed:
Broth:
- 10 grams Agar Agar powder or flakes. (Note: Some of the food grade Agar on the market has preservatives in it, the best results will come from a quality preservative free Agar, examples of good brands are NOW, Tic Gums, or Fungi Perfecti Agar)
- 10 grams light malt extract.
-500 ml H2O.
Plates:
250 ml (8oz) Ball 'elite' Mason canning Jars.
Wide mouth synthetic filter discs.
(If you don't have filter discs you can use Citric's self heal lids and they work just as well, Just make sure to drill a third hole to prevent overboiling in the PC )
Wide mouth canning jars lids with (2) 5/16 inch holes drilled in each one. This keeps the agar from over boiling while it is being pressure cooked.
Wide mouth rings.
Cooking the Broth:
Put 500 ml COLD H2O in a clean pot
Add 10 grams light malt extract.
and 10 grams Agar Agar flakes.
You are going to want to start off with cold water and add the ingredients, then bring the media to a slow simmer, the broth should look slightly chunky at first but it will melt after a few minutes of stirring.
Simmer at a low boil for approx. 3-5 minutes or until the majority of the flakes have melted.
NOW CAREFULLY POUR APPROX. 60ML OF YOUR AGAR BROTH INTO EACH JAR
Assembling the Plates:
To assemble your plate first you lay the lid down bottom side faced up on your jar.
Next you lay the filter disc on top of the lid.
Screw on the ring firmly, but not too tight.
Cover your jars with foil just like when you make grain or PF jars.
Stack your jars in your PC with hot tap water just barely coming above the liner insert, about 5mm above the bottom of the jar.
Finally, PC for 45 minutes at 15 PSI and let the Jars cool & set over night.
The following day after letting them cool overnight in the PC you can inoculate them in a glovebox or in front of a laminar flowhood. This is what your final product should look like after you add your favorite mycelium!

Couple questions since I'm about to do something very similar to this tek. First instead of using filter discs I used plastic lids and melted 3 1/4" holes across the lid. I made the middle one into a silicone injection port and placed micropore over the other two. I'm wondering if this will suffice for fae and prevent any boil over or any other reason my idea won't work. Also a lot of ppl preach by fast Fred's and your recipe is slightly different so I'm wondering if you've experimented and found yours better or if I should follow fast freds my first time. Also since I'm unable to measure each one precisely how thick would u say I should aim for in the middle of the jar?

You need to punch in the words 'elite' ball mason jars when searching for them. You will want the 8oz ones for Agar work. I actually use the 16 ounce ones for exotic side projects like Zappos, etc....

I used to use these ball elite jars, but they have a convex bottom, but I can pour ten flat bottom Tupperware containers with 500ml of agar media, they have identical diameters. How many you get really depends on how much solution you pour in and how big the jar/container/petri dish

That depends on how much agar you make and how many jars you have.
If you follow the measurements in the op you should be able to get 8 no problem out of 500ml agar. You may even be able to stretch it to 10 if you pour a little thin.

I don't use jars these days. I just made the tek a while back to help noobs out.
I still use the elite jars though. I use those and the full pint jars for exotics. They are the perfect size for countertop grows.
These days its always disposable sleeves of 20....

I tried this tek a couple times now and every time i go to open the jars for inoculation, there is tons of condensation that pools up in the jar like an ocean of excessive h2o. I got the jars leanin on their sides so the water just pools up on the side and doesnt hit where i place my transfer.
Any idea why this happens? Im about to try it with 300ml water instead lol. Im using agar agar 'Eden' brand and it says 'vegetable sea flakes' on it.

It could be a couple things. How loose did you leave the lid in the pc? If it is too loose it can draw water in the form of steam into the jars while the pc cools.
Or it could be a little too much water to agar ratio. I have had that problem but always just poured off the extra water if it happened. It usually never happened with all the jars though.
You may also want to try and find a better agar. The eden brand flakes have a couple downfalls to them. The one that comes to mind that turns me off to using it anymore is the fact that eden agar has food preservatives in it. While this is good for extending the shelf life of their product it is not so good for mushroom mycelium.
Plus the higher grade agars gel up a lot better. I recommned Tic-Gums agar. It is the best I have worked with and I have tried all the best ones.

Well ive noticed that the water is coming from the bottom of the lid which drips down the sides. So ive come up with a solution to PC filtered lids seperately and use normal lids on the agar jars. When PC cools, im gonna unscrew the ring of the agar jar, and quickly replace the normal lid with the seperate PC'd filtered lid which doesnt have water condensation.
Where do you get the tic gums brand?

It is pretty pricey if you get it in a small amount. You have to get it in bulk unless you get it from their chefs section.
You want the 'Agar RS-100'. It is a special agar that does not require boiling. All you have to do is heat it to 180 degree and it melts completely evenly throughout the liquid.

I just made agar yesterday and I tried this no-pour thing. I figure it would save on time and reduce on contamination if I get it right.
We'll see how it goes.

Yea, this works well too. Just make sure the polyfil isn't too far down into the jar to the point where it is touching the fluid or it could cause problems for a couple reasons. It may act as a wick and you could lose some or all of the media from the jar and dump it into the PC or it could cause contamination problems if the polyfil gets soaked in the nutrient enriched media.
You can also inoculate through the polyfil or drill a 2nd hole and make it into an injection port.

Sorry to bump an old thread. I'm new... and I'm wondering why you suggest a third hole if using a self healing port. I'm thinking of doing a hole with straight RTV and a second hole (11/64") with EZ Felt. Would that be at risk of boiling over? In another reply you mentioned having a polyfil hole and then a 2nd hole for injection = just two holes. Thanks for this post!

Thanks for the write-up. I'm still a noob and still learning. I keep ending up with contam in my agar and then my grain jars. Hoping this will help take care of both.
I couldn't easily find the 8oz elite so I picked up the 4oz clear Ball jars from Target. It doesn't give me as much surface area on the bottom, but I think it will work fine. I may regret it; We'll see.
I used 2 layers of micropore tape instead of the filter.
I didn't make an injection port because I'll be doing agar to agar transfers mainly for now.
Any strong opinions on these modifications?
I do have one idea.... Let's say you have a jar of agar that develops contamination. Could you just throw it right back into the PC and re-use the agar? Seems like a really easy way to clean up and prep all at the same time!
