Liquid Culture Recipe - Does this sound about right to you?

About to make a LC media. Just want to double check with everyone here before I make it. Here is the recipe I will be using.
10g Dextrose 10g Light Malt Extract. 800ml Distilled Water.
Does this sound good to the rest of you?

Looks fine. Typically you want your sugar sources to total about 2-4% of the final weight/volume. Yours is about 2.5%.

I do by volume. 1 tablespoon of each per 1 quart/1 L Filtering (a few times) as I load it into LC jars + stirbar PC with lids loose for 20-25 minutes with a prior 5 minute "off-steam" PC cools, tighten lids. Let cool for 24hr-ish

Yeah I was reading that "By Volume" And I converted a recipe I found here on the board Agar put on (I believe it was agar) about a good mix for LME/Dextrose LC and he wrote out the same you did... 1tbsp Light Male Extract 1tbsp Dextrose 1 Quart water
I am just not so certainly familiar with Quarts and Tablespoons ect... I prefer the metric system for clear amounts and accuracy. So I weight out 1 table spoon and rounded to a even Number and got 10g. for both LME and Dextrose... then poured out 1 quart of water and got 720g or rounded liquid measurement 800ml. or 760g of water
that is where is got the recipe:
10g Light Malt Extract 10g Dextrose 800ml Distilled Water
I was am just trying to confirm that this was a accurate representation of the 1 tbsp lme 1 tbsp dex. 1 quart water.
one quart would be 800ml roughly right?... Maybe I should use 1000ml or 1 liter of Water to dilute it a bit more. so It is not so strong I have read less is more when it comes to LC

Hey man... your the shit. Thanks for converting my concentration level for me... This is what I was most concerned with.
So just to make sure I understand what your telling me... I have approximately 2.5% dissolved sugars in my Media per volume of water. This being the Light Malt Extract and Dextrose.
And the Ideal amount of dissolved sugars wanted is between 2% and 4% the volume of water.

Hmmm. I tell you what buddy.
You write the TEK and I will give it a try. I have all the materials for agar. MEA Agar and PDYA Agar... Drop me a PM when you make it and I will try it and post results giving you credit and proving your TEK for others to try....
Sound like a interesting way of making LC Media.
*Ding* just finnished brewing my LME/Dextrose LC Media... now time to PC

Thanks.
I use a syringe to calibrate my scales. Or a coin that I know the weight of. UK pennys are 3.5g, and demonination of dollar bills are 1g.
Put empty syringe on scale, zero(assuming its digital) add x ml of h2o, scale should read xg.

Sounds like I should get on that... because I just mixed a batch of LC and the recipe i just mixed could be all screwed up

Hmmm thats... I dont know?
I tried 1ml, 5ml, and 10ml and the scale said 1g, 5g, 10g... Maybe the scale starts to become less accurate the heavier of an object thats placed on it.

I am really not worried about the scale anymore after the test you just had me do. I have measuring cups scaled in ml... so I really have no need to measure anything heavy. like water I have just been using it for 20g and below measurements like LME Dextrose Agar-Agar and the lot... Now I know its good for the small measurements I am sure that batch of LC I made earlier is all good.

2-4% sugar content for lc and agar. I prefer 2% on agar for faster colonisation. 4% for lc, for stronger lc. Thats just me though.
I also do it by weight, not volume.

I'll take a crack at that...
1 tbsp = 10g Dextrose 1 tbsp = 10g LME
And I have a 2.5% mix say's MycoAu
Quote:
MycoAu said: Looks fine. Typically you want your sugar sources to total about 2-4% of the final weight/volume. Yours is about 2.5%.
So based on that... You would do some math. (here I will do it for you... because I am awesome.)
2.5 - 4 = 1.5 so add that to 10g and you get 15g
-----------------------------------------------------------------------
So you would want to mix this...
15g - Light Malt Extract 15g - Dextrose 800ml - Distilled Water
and you should have a 4% Liquid Culture Media. (provided that MycoAu was correct saying I have a 2.5% mix)
Did that help?

two tablespoons it is.
Jw, because i've been having fucking bacterial contams RIGHT at the end. Maybe the high sugar content will make the myc stronger ? I've only heard good things about 2:2 lme/dex, now I know its ~4%

Wow... I got all excited thinking I am fucking smart... I didn't even answer your question... so here is your answer...
If: 10g = 1 tbsp then: 15g = 1.5 tbsp
so you would mix this
1 1/2 tbsp - Light Malt Extract 1 1/2 tbsp - Dextrose in 800ml water or 1 quart (approximately)
Okay I think I may have helped... this time...

Negative Ghost Rider...
If what MycoAu was correct in me have 2.5% media then you adding 2:2 you mean you would have a 5% solution...
you need to add 1.5:1.5 per 800ml water... to have 4%

Ohh your going 1000ml of water... okay then yeah that would be pretty close to 4% then...
I was basing this off my mix using 800ml of water...
okay... okay... you right then 2:2 it is
1 quart is 1000ml ? (see I dont know standard american measurements I always use metric so i dont know really what a quart is...)
