Top-notch thread Stonesun. More folks should cultivate these, my fave. Never had a succesful jalisco culture though...just tampanensis and mexicana strain A.

I do have a question and you seem like you might be the guy to ask:
Can I clone sclerotia by adding directly to an LC? I have some Mex A jars about 3 months old now, popped one the other day and found 120ish grams of sclerotia, nice. I was gonna pop another soon and wondered if it would be worth putting a sterile, fresh stone into an LC or 3?
I have tried G2G before with these with poor results, I think they are less forgiving contaminant-wise than cubes due to the extended invitro period.
I have a few agar plates left but wanted to use them for another project. I'm guessing it'll be xmas or New Year before most places are shipping reliably and I can get more, especially with the weather over here..
I read the whole thread (and several others) but can't seem to find this mentioned, any thoughts??

I am betting that agar would be a better place to start.
There is no guarantee that sclerotia are clean.
You could try washing it briefly in H2O2.

I'm almost certain agar would be a better place to start too... However, in the abscence of available dishes, I might give it a shot unless someone has tried a few times and knows that it's a waste of time...??
I was thinking of removing a stone from a clean jar (pre-harvest) with heated tweezers and just dropping it in to some LC. I have plenty of stones available and no shortage of LC jars and honey. I think I'm gonna give it a whirl, will post results when I'm done.

I do this all the time, it works great. Why wouldn't sclerotia be "clean" if you pull it from a sterilized jar of sterilized substrate?

Not so. They are actually VERY forgiving. I've harvested sclerotia with my bare hands and separated the colonized rye. Threw the grains in a cereal cup (didn't even washed it out), cased with 50/50+ and it produced just fine without any contamination. This being said I will never recommend the "Neglect Tek" to anyone!

I don't think I was being clear there.. I meant that G2G is more tricky as they sit for so long, althought the myc may dominate; if there's a single contam present it will probably show itself in that time period. I've had jars turn bad after 4 months fully colonised. I like to use LC or agar as I think the risk of airborne contams is less when transferring, not a big G2G fan really.
Cool, glad to know you've done it successfully, thanks man. I know the sclerotia will be clean, my concern was more whether it would produce myc easily. I will get started on my LC's now, hopefully silencing some doubters along the way.. lol (You know who you are!! )

This make sense. Thanks SS.
I suppose that grains from a grain jars are cleaner than a sample from a birthed BRF cake. I tried an LC from the latter without success, but then again I was just getting started.

Some of my stones turn almost a blackish brown color, Not like the normal stone brown imo. What would you say that is? I wouls suspect bacteria?
Thats happened in nearly all my jars, from 3-6 months old.

Pics please! I definitely had some pretty black streaks through some of my healthy ones, usually against the glass it seems, I'm sure you know they can do some funky colour schemes.
All my late-stage contams have been white speckles that form on the stones and substrate. To be honest I'm not 100% sure mine were contaminated, they were between 4 and 6 months and just weren't quite right so I tossed them. This has only happened to me with G2G which is why I avoid it.
