Do Azures require agar?

I'd like to do some Azures. Will I have to grow out mycelium in a petri dish? Somebody told me in another post that you could germinate spores on cardboard. I didn't realize this was possible.

Is it?

I dont think you have to start azures on agar.
But if you do i'm about to try cardboard squares on pda + peroxide agar.I think the idear is that the spores germinate on the cardboard(were other spores find it hard to germinate)then the mycelium moves onto the pda + P agar which helps keep contams down.
You can also use cardboard and beer theres a thread here somewere about it.

Hi!

I started to isolate spores on Agar. Then transferred onto rye (mixed with some wood chips and cardboard) and then onto wood/or cardboard.

Forget to put agarpieces onto cardboard it won?t work. That?s my opinion.

yes to both. the pic i posted is azure myc germinated on MEA. the one posted here below shows spores germination on cardboard placed on a PEROXIDATED agar plate

here is the same petri from the previous post, after the spores have germinated on the cardboard (slowly) and then grown over the - PEROXIDATED - agar (aggressively)
  note that this tek is used to benefit from low contam rate of cardboard (as opposed to non perox MEA) for spores germination. yes, the cardboard i used with success is the thick gray cbd (from the back of note pads) :smile:

mycelium seems to prefer gray cardboard, like the backing of a notepad .not sure what the difference is .egg cartons work good too .

Do you still have to use a flow hood, or glove box when using cardboard, and peroxidated agar?

Do you have to PC anything?

I use peroxidated agar and never have to use a flow hood, or glove box. I just clean up where i'm working.

The process for making peroxidated agar is the same as any agar - mix, and pc - but in the end, you add some peroxide prior to pouring, I use 4cc's in a 500ml mixture.

You will not be able to germinate spores on this agar, but as far as cloning and stuff go, it's great!

Do you use the standard 3% H2O2?

Could I buy a jar of presterilized agar, and add peroxide to it?

I won't be using it all the first time, will it keep? Will it have to be refrigerated?

So I pour the dishes, and let them set, then take some cardboard soaked in boiled beer, and stick them in the dish, then streak some spores onto the cardboard?

I think I've almost, but not quite, got it.

I?ve been using a different method on various occasions, and it worked well.

I make a fairly diluted spore suspension and inoculate a classic PF cake with it on 4 spots with little inoculant.

One or more of the spots usually show clean growth, then you can transfer those spots to either peroxided agar, or directly to peroxided grain.

If you buy pre-made agar, yes you can just liquify it in a water bath, then add the peroxide. And if you want to keep the agar for later, you can just recover, and reliquify when ready for more use (you will need to add more peroxide at that point, and your mixture might become a little bit watery (some water on the top of the media) but it should work, even after the 1st, and if not - then you can always just try and be as clean as possible, as you would w/o peroxidated agar. It's not that easy to make a mess of, if you're careful.

I would imagine you could go to cardboard, and from there to chips. I believe stamets talks about cyan bed prep that way in one of his books. Cardboard spawning chips, long sheet of colonized cardboard with chips on top.

it would certainly help (to have a flow hood or glove box) but its not absolutely necessary. pc would be necessary for sterilizing grains to be used as spawn
