Jars not colonising :(

I inoculated my jars about a week ago now, and still don't see anything!

I sterilised them, did everything properly, have kept them in a plastic container at the right temp...

Wahhh :'(

What could the reason be?

More information is required.  What type of jars?  Are your inoculation holes covered? What temperature are you colonizing at?

Where did you get your spores from? They might just be bunk spores...

But more likely, did you allow your jars to cool for 24 hours after you PC'd them? If you inoculate a hot jar, the temperature will kill your spores.

Grains or cakes? What kind of filter? What temps? How many jars? Is their excessive moisture?

You don't need them in a plastic tub.  Can you open a jar and smell it?  A sour or old sock smell is a sign of bacteria.  No signs of growth in 7days means something is wrong. ~Pixie~

I covered the jars (each with a hole in) with 2-3 layers of foil, secured it tightly.

I didn't cover up the inoculation holes after I inoculated them, as it had dry Verm ontop.

There's no gas exchange that way. Dry verm on top of the inoc holes? Never heard of anything like that...

No, I mean the jars are 3/4 full of mix, and the last 1/4 is dry Verm.

Air tight jars will stall. Haven't you watched the videos? What are you doing?... What are you following?... Remove that foil and what type of filter?... Who do the lids look like? The verm layer is there to stop contams from attacking your mycelium or substrate.

The verm acts as a filter for the air, it should exchange air ok, sometimes near the end of colonization if it seems like it is stalling, you can turn them upside down and what happens is when you do that the mass of everything comes down and creates a vacuum effect and brings more oxygen inside the jar to finish up.

I must admit, I sort of rushed things...
I still have half a syringe left, might have another go.

Also, is it normal that the syringe had clumps in it? It looked clear apart from the clumps. (I shook it up as much as poss)

The jars I used have screw on tops

well first thing take that foil off..its only used to keep out the water while p/cing. how many holes in your lid? how deep is your dry verm layer? But honestly, It sounds like you need to go watch rr's videos.

there should be four holes when making cakes to inject in four different spots and i usually put cloth band aids over mine. dont quote me but what would somebody else recommend covering it with a coffee filter and rubber banding it after your done? just an idea i really never did cakes so its not my expertise

A coffee filter is quite useless for keeping contams out. The verm layer is for keeping contams out.

Bullshit! This is a tried and true method  to renew growth to stalled jars! Also it's not for CO2 drainage as you said it simply allows the cake to settle this give a brief exchange in gas. I don't give a shit what someone thinks they know if it works and has worked thousands of times you can't dis it!!! It don't do shit to the verm layer that is pure crap!!!! Unless someone doesn't have a clue and tries to shake the damn jar then they deserve what they get. Myc generates heat regardless if the cake is right side up or upside down and that doesn't always take care of gas exchange it simply pushes out gas from the top of the culture.

yea dont cover it. Just take the foil off and losen the lid to provide g/e. was the one whole in the center? Oh and your left over spores should still be good. Just be sure to flame sterilize. Are you gonna watch the vids?

The hole is in the center, I've removed the foil, loosened the lids, and put them back.

I see myc in 2 of them at the bottom (although one of them has some wet bits...) :frown:

If your syringe has clumps in it, go ahead and dilute it.  You want to be inoculating with fairly "light" syringes anyway.  A syringe with clumps of spores can make a handful of syringes. 

If you're really low, you just use less inoculant.  I inoculated a quart of grain with less than 1/4cc; just colonizes a little slower. ~Pixie~

give it 5 weeks. I have seen spores take 5 weeks to show first signs. People are too anxious and after a few minutes or days after inoculation, they panic and think it is a failure. just sit back, as long as there are no contams, let it go.

I flame sterilized the needle when I did it the first time, I heard popping - the sound of spores dying??? :'(

Also the needle turned kinda black so I cleaned it off / cooled it with a Dettol wipe, is that ok?

My syringe made popping noises too when I first inoculated, I think it's just from verm that gets stuck up in the needle and heats up and shoots out.

Ya you can use the syringe again just flame the tip or clean with alcohol....don't re-inoculate them...just be patient...

Certainly sounds like what happened to me with air exchange. Eventually i lossened up the tin foil lids a bit an viola! fully colonized in 3 weeks waiting for pins.

You'll have bigger yields when spawning to bulk and applying a casing layer but I'd dunk and roll if I was you. Do the previous for your next grow.

i followed the tek exactly and watched rrs vids and same problem...got new spores and tried the old formula so maybe that will work
