Just to clarify...

When I go to prepare my slants. I want the leading edge...now, if in the case of a petri dish that has grown out to the edges of the dish as opposed to half-way, does that count as the leading edge from the original inoculation or do you drop a new piece into a new dish and go with that edge (which seems counter-productive to me).

I'm kind of a noob - but I have seen lots of petris contaminate - so here are my thoughts:

The reason to get it far from the center - you are hoping the myc. will outrun any contamination on the original sample.

Also try to get it before it reaches the edge; because any contamination that seeps in under the lid usually happens around the edge. This is something I read in a Stamets book and it makes sense.

If it has reached the edge and you want to give it a try anyway - I would take a sample from halfway in.
Furthest from any contams in the center, and the edge.
