 Kick-Ass LC Method I Gotta Try (Updated With Pics of an LC)

This is RR's LC recipe. I am just posting it so that I can find it easily under "My Threads". I don't want to lose track of where I found it.
Colonize a quart jar of rye berries with your species of choice. Shake well to break up the individual kernels of grain. Allow to recover for a day or two just until the kernels begin to turn white again, and then add a pint of sterilized water to the jar and shake well. Pour this water into your flask and add up to three times the volume of sterilized water and allow to stir for a few days. Fill your syringes and use. You can store this water, even in the syringes for at least two years in the refrigerator and use whenever needed.

If you search "Grain LC" you should be able to find it. It is really one of the better ways to do LC's without a doubt.

agar has one also, there all the same principle. but i am curious on this one, does it use syringes to inject the sterile water into the quart or what? i only have small syringes and when i do this it is alot of trouble using 10cc syringes to inject sterile water into the quart about 10 times to get enough to strip the myc off the grains and then doing the same to get it out and into the jar of sterile water. i know i just need to break down and buy some larger syringes at the feed store. But do you think its possible to do the above with out the syringe in a glove box and have it work with out contams?

It's def a great way to do it but ya probably want to do it w/an isolate so why not just keep working off that solution into a larger batch and skip the Rye completely?MS ok though I guess

sure that would work in a glove box (I think)
I would just go buy a bigger syringe because the feed store is like 5 mins from my house.

It should work fine if your clean enough to keep it uncontaminated,clean room,still air, gloves and alcohol..You know the drill

I use a flowhood, so I simply lift the lid off and pour the water right in from jar to jar, and jar to flask. By the time you guys get into bulk growing where you want that much LC, you really need to pony up the dough for a flowhood.
Mostly, I just use that tek to make syringes for demonstrating the brf tek at schools and such. For my own growing, I do grain inoculations in quart jars from agar wedges, and when those are grown out and tested, I do grain to grain transfers into filter patch bags of rye that hold 6 to 7 quarts. Each of those rye bags will inoculate 20 more, so I can expand mycelium WAY faster that way than by using LC. Syringes are for kids.

what about senescence?
how often do you run into that when doing g2g into 6-7 quarts, then to 20 more?
that sounds like it would have a bit of an issue.

Yeah, I dig what you are saying. I have started to do some isolating, but not in earnest yet. I got finve isolates off of an Ecuador strain just to see if I could do it. I am going to isolate in earnest though, off of some pan cyans.
I actually want to use this method to make LC with my edible strains/medicinal strains. I have plenty of solid cubes around for my personal use and am starting to grow shiitakes and oysters. I am going to make some master culture slants to save them for perpetuity. But I would like a solid way to make LC because they colonize rye so fast. Since a guy eats a lot more shiitakes in a sitting than cubes, I need a lot more jars of rye.

Well, never mind that last post. This method seems a lot better for bulk growing. God, I hate being so broke right now. I wish I had the bread to build a flowhood. I am sick of being in school.

Yeah, i hear what your saying RR. but some of us are feeling the decline in the economy in the US. I have been trying to build a flow hood for awhile now, i got the blower and the filter's i just need to get the wood for the box and a few other items and ill be on my way. And i was just speaking in another thread about needing and wanting to get into doing spawn bags more. I already use grain to grain often, i like LC better for some reason. but when i start to use 6-7 quart bags ill probably be using g2g more.

So I had two extra quart jars of rye fully colonized with shiitake culture sitting around, and I used this LC TEK with one of them. The only thing different that I did was to also add some LME to the sterilized water. That was two days ago almost to the hour; and look at it now. (The pic is actually pretty big if you click on it).
I will never go back to making an LC any other way.
P.S. I also sterilized two bags of sawdust supplemented with bran and gypsum. I used my other fully colonized jar, and then I put the one I used to make the LC into the other bag. I don't see why it shouldn't pull double duty for me.
