Poly-fil sterilization, prolly a repeat

Probably been answered before, but I couldnt find it in a search. How do you sterilize the poly-fil for an air port syringe, is dipping it in alchohol enough, or does it need to be PCd? Ive heard of both, but what the hell works the best?

From what I understand, polyfill doesn't harbor contams. I would just put it on your jar lid, cover with foil, and sterilize in the PC. Make sure your syringe is sterile though. Are you making LC?

Yeah, i actuallyalready made 4 of the baby bottle LCs just to get a feel for it, and i can't find out which rubber hose will withstand pressure cooking for Agar's LC tek. Its been like 2 weeks, and there are thin whispy strands of myc, but nothing like i see in pictures, and i think some air exchange would speed the process and make the myc grow. So I read about taking the needle off a syringe, stuffing it with poly-fil and sticking it in the self-healing injection port to allow oxygen in. Someone said simply dipping the poly and needle in alchohol was sufficient but i dont want to risk losing these LCs due to inadequate sterilizing, and i don't want the poly fil to melt in the PC, so i'm not sure what to do.

Polyfill can withstand PC temps just fine.

I use it all the time in my grain jar lids and never had a problem yet.

I cant really help you much with the LCs since I dont use them. Instead I use a completely colonized grain jar, add sterile water, shake like hell, then suck the water back out into a syringe.

Does anyone know if an alchohol bath will do the trick, it would be much easier, lol

I'm thinking that it would be fine in the PC.  I'm going to have to agree with Alkaloids about making sure to cover with foil though.  Good luck Rowdy and let me know how it goes.

So, i PCd one last night, and yesterday afternoon, i soaked one in an alc bath and stuck it in the LC that I had extras of, so they will be my tests. We'll see.

I did that to three LC jars which I screwed the lid (sans any holes for air exchange) down too tight while PCing and ended up injecting an entire syringe worth of spores into one jar on account of the extreme negative pressure.
Solution:
Take the plunger out of the syringe (keep the needle on!), stuff it with polyfil, wrap in tinfoil and PC it. Remove from the foil and flame sterilize the needle once you are ready to equalize the pressure.
I don't think you need FAE while growing a liquid culture.

Bubba, have you had any contamination issues with your syringe making technique?

Hey Bubba when you use the pollyfil how tight do you roll it up?  I don't want to roll it so tight that air can't pass but I don't want it so loose that contams find there way in.

On rare occasions when the jar has a bacterial contam and I dont notice it, I have had a few bad syringes. Just make sure your grain jar is healthy and you are sterilizing the needle and water. You shouldn't have any problems.

For a quarter inch hole, I grab a piece of polyfill roughly the size of a cotton ball, fold it in half a couple times to wad it up and push it through the hole before sterilizing.

As long as the hole isnt packed really tight gas exchange will occur.

i use latex tubing found at homedepot kinda expensive $20 for like 10 feet works great i use agars lid with the 2 tubes commin from the top stuffed with polly NO VACCUMM!
