Ok so your supposed to flame sterilize your needle before inoculation....some say to flame it and wait a second or two...some say flame it and let it cool for several minuts...some say to flame it then use an alchohol wipe to cool it and to further sterilize...some say flame it then squirt a drop or two to cool it then start innoc....So wich is it...also "IF" cubes and spores are extremeley temp sensitive (ie. incubation temp no higher than 86 ) then after flame sterilization shouldnt you let it cool for like 30-60 min....aarrgghh so confusing

86 is max ideal growing temp..

the thermal death of spores is much higher.

I flame, alcohol wipe, and inject one jar, repeat.

If the alcohol doesnt cool the needle.... the spore solution will.....

Wrong.  Any MYCELIUM temp above 83F slows growth considerably with P cubensis.  The 86F figure is flat out wrong in TMC.

Since we know that mycelium produces heat as it grows, it's important to keep the ambient temp below 83F for best results.  For Petri dishes, keep it at or below 83F, but for pf jars, keep it below 81F, and for quart jars of grains or bulk substrates, keep it below 78F to keep your mycelium in the correct temperature range.

As for a syringe needle, heat it red hot, and then use it a second or two later.  It's counterproductive to wipe a red-hot needle with ANYTHING because you can't get more sterile than red-hot.  The first drop or two of spore solution will cool the needle, and the rest will flow perfectly fine into your jars.

Flame until red-hot between each jar to prevent cross contamination.  There is no need to do anything between holes of the same jar.

god, no matter how many times ( sometimes many times a day) you post about incubation temps the 86f misinformation still pops up.

Also just for the sake of argument i leave my incubation temps at 80...I was only using 86 as a nearing thermal death temp "nearing" wat is thermal death anyway 98-109, but more importantly what is the thermal death of cube spores

The thermal death points for most bacteria, molds and spores during pasteurization  are as follows, 185F - to 212 F, when heated for a few seconds; 180 F for 2 minutes; 170 F for 5 minutes; 140 F for 20 minutes. Even some thermolphilic fungi and it's spores can only survive up to about 145f.

Dry mycelium may be able to withstand high temps..i know of no studies done.

As the mycelium loses water its ability to withstand cold and heat is enhanced.
This is how myc can withstand freezing temps.
