agar in 1/4 pint jars

I'm planning on doing agar in little mason jars rather then petri dishes. My question is, Can I put a 1/4 inch hole in the lid and stuff with polyfill for clean airflow? I am pretty sure I know the answer but just incase somebody knows something I don't please let me know.

Absolutely. Personally, I like these jars for agar work:



They're 1/2 pints, but they're also wide and really short. I drill two 1/4" holes and use postal tyvek as a filter for a clean look like so:





They're perfect for dropping pins in for cloning purposes:

I used to utilize baby food jars as well...with the lids on tightly(after autoclaving).  I think such a small quantity of mycelium does fine without the need for additional air exchange (most species).  My adult friend would eat the baby food for me (yech). 
I incubated my petri dishes(or baby food jars) in ziploc bags for many years without any problems, and without reopening them until required. Now I provide them weekly air exchange(open bag and close) unless they are in long term cool(or other) storage.  But you can typically get away without opening them until needed.

In my opinion petri plates are better, for storing a culture is necessay a vial or a test tube.

Using jar or little jars you use more medium and you work bad.

If you don't have the money for plastic petri plate buy 20 glass petri plate one time and they last a life.

Plastic petri however are better because with the years the glass one aren't so transparent as new.

Using jars, drops of water inside the jars can be a problem, and cause bacteria contaminations or slow the growth of the mycelium.

I have had zero problems using jars. I also use pyrex petri dishes and find that of the two, petri dishes are far more likely to have condensation than jars. Another advantage to the jars is that you can inject a spore or culture solution into them without the need for a glove box or flow hood. Yet another advantage is that they can be sterilized with the agar inside of them without worrying about the agar boiling over and making a mess. That's much easier than having to pour hot agar into plates, which again requires a glove box or flow hood to minimize contam risks.

The pros for petri dishes (IMO) are that they are cheaper (plastic ones anyway), they take up less space, they are a bit easier to store and stack and you can see through the tops of them, which makes it easier to see what kind of growth you have.

i want to try the method of the jars another time and see what happens with the condensation. However the petri are really more useful than jars, speaking about space and way to work.

I have strains stored in baby food jars and they work fantastic for that, but I wouldn't recommend doing any kind of isolation work or cleaning up of cultures in a jar. Petri's are more suitable for that kind of stuff. All you have to do is keep your dishes insulated to minimize condensation.

I just those jars with great success.  I put four nail holes in the lid and then use tyvek between the lid and the jar.
