Same here tripsis ;(
I made about 5 clones, non of them survived - same story as yours!
Then i made spore print from shop button and as you know sterility in shops is 0% so around most spores became contam! Other spores started germinating only after 3 week :O :S
Well maybe that the secret of comerical shops for shrooms xD

You actually got button mushrooms spores to germinate? Wow, that's supposed to be really difficult. I took some fresh prints so I'm going to put some spores on agar this week. Hopefully I'll be able to get them to germinate too.:)

tripsis,
Maybe you should try a different agar medium - Say if you used PDA then use some MA or MYA.
I once got some button mushroom spawn (a few grains) and tried it on PDA - Very very very slow growth....
The agar plates dried out before the culture could get going properly.

what was the source of your starting material? (ex. spores from xxx, culture from xxx, ect.)
theres probably not a whole lot of agaricus cultivators on this forum I remember reading posts of RR saying he couldn't get his to fruit when he needed them for a video.. that made me not want to try growing agaricus mushrooms.

How long have the chunks of mushroom been on agar?, sometimes bisporus takes 10 to 14 days before you see growth.

Yeah, I posted a bit prematurely. It worked in the end, not all the pieces of tissue, but several. Still have it all on plates though, growth is so slow. I'll start some jars of spawn tomorrow though.

I don't see how it would make a difference how the tissue was taken from the mushroom. As I said, it did work out, it's just Agaricus mycelium is very slow.
I'm not keen on the idea on cloning straight to liquid culture. I prefer cloning to agar to ensure the culture is clean, then proceeding to LC later.

tripsis said: Hi, I have been growing some button mushrooms. As I want to continue this I decided to clone off one. I did so yesterday and placed a small piece of tissue of several agar plates. Unexpectedly though, the tissue has turned brown and appears to have died.
My question is, is this somehow due to senescence of the strain because it is commercially grown? I tried cloning off shop bought button mushrooms with the same results, but I attributed the failure to the fact the mushrooms were days old. This one was most definitely alive when I cloned it.
Is it normal for it to go brown like that and can I still expect to see growth at some point?
Thanks for any feedback.
I have tried cloning some other mushrooms on agar before and quite often, the tissue would turn brown and soggy and look dead but after a while, mycelium will grow from it. Sometimes however, it will not grow mycelium. Just my personal experience.

Yeah, it was quite strange actually. Many of the pieces of tissue that went brown, ended up showing growth, while the pieces that stayed firm and white did not.

My button mushroom clone didn't work either (on PDA), since it is a dung lover I'd suggest sterilizing some horse manure in a jar and throwing your tissue clone in there.Experiment with water content for best results.
