 Volunteer fruiting! Shiitake

After many many plates and transfers I received a nice surprise. A strain of mine # 102 just produced a nice volunteer. Thought I would share.
And the same in broth. It grows very nice and thick.
Spawn has already been made and will be transferring to sawdust/chips this week. Wish me luck!

I sure hope so! We will see in a few weeks. Got a couple of tents up and a cooling unit for it. I have managed to get the tent temps in the stable 60's so I will also be fruiting the shaggies right before the shiitakes take. I dosed the shaggies with lots of nitro in dishtubs full of sawdust,alder chips,newspaper,coir,and blood meal topped with 2 inches peat casing. They took to the mixture like mad. Just another project I have going.

I am really tired from work so I am going to do a numbered list.
1- Spore print
2- About 5 plates
3- Streak spores with loop
4- Dipped the loop into small amount of sterile water
5- Sterilize loop and cool
6- Dipped loop back into the spore solution
7- Streaked across plates
8- Isolated localized sectors as best I can and transfered them into about 30 plates
9- Made another 40 plates and isolated any remaining desirable sectors again
10- Repeated if necessary
11- Selected best cultures with most even vigorous growth
12- Watched for cultures that exhibited the best consistency. and tested those plates on separate plates.
13- when I found what I believed to be the best of the bunch, I numbered each good culture and made backups.
14- Made new plates of each, made an lc of each, and 5 grain jars of each.
15- Watch carefully for anything undesirable or desirable.
16- Keep watching.
17- Found a few that are very aggressive and form aggregates profusely. Some had a few large aggregates, some small, and some with small and large. Matching cultures exhibited the same and were consistent. I was very happy with the results "Thats when I saw the plate that produced a volunteer". Even happier! Time to make sawdust blocks.
18- Transferring grain into bran sawdust bags this week.
19- Tents are up and time to test selected cultures for fruiting ability and record results.
This is probably a little crude compared to others, but balancing it with my work is quite a chore so I do what I can.

no doubt you did the right things. but, transfer tissue from agar to agar, could select weak strains, isn't? i read that 3 or 4 transfers are the maximum the tissue resist, and i have very dissapointing results with ONE lc to lc transfer

Your running with the mycelium! Your not letting the culture grow out across the dish every time. You do try to keep the cell count to a minimum as you work with it. You do what you can with what it presents. And it's different every time thats for sure.. Weak strains are easy to identify! They are very wispy and can be distinguishable next to better ones. I go for the faster more slightly rhizo types which grow more linearly, at least for the shiitake that is. They seem to give the best results.

The three transfers 'limit' is for grain to grain transfers where the mycelium is expanding exponentially. In a petri dish, this doesn't happen. The flat plane of the dish limits growth to the surface, greatly reducing the number of cell divisions. Also, as Lipa said, we don't fully grow out the petri dishes. We transfer when the mycelium is 1 to 2 cm in diameter. We can easily make three or four transfers in a two week period, isolating a strain or cleaning up a culture from contaminants.

It only means that It could be "quite likely" a good fruiter. We won't know until It is fruited on the blocks. I will keep you guy up to date on how it is doing.

i'm growing an lc from a shiitake which fruit in paper, in order to view what happen in sawdust. could be a strong strain, isn't?

You fruited shiitake on paper? Wow! I want to see that. That would be cool.
I dumped one of the grain blocks from that dish into the chamber while the sawdust is going to see what would happen. There is primordia on them and they are gently getting bigger. This is a good sign. I cannot wait to see the blocks go off.

i run the entire process thinking was reishi  no cold shock, no 24 hs submerged in water. they aren't "the" fruits but grown in paper! probaby sawdust give interesting results.

Here is a pic of the #102 strain starting to fruit on rye grain. By the way, the #100 and #110 are looking pretty good too. The only difference is the # 102 is taking longer to colonize. #100 seems to be the fastest, but that is not always a good thing. #110 Buckled the quickest.
Sawdust blocks are next. Soon I promise.
The quarts on the right in front of the bags are the # 110 running down through oak sawdust/bran They were inoculated on the 15th of this month.
