total isolation of a strain

I'm looking to isolate down to pure sub strains within the cubensis species. wut I am wondering is this:
Is it possible to isolate a sub strain down to a relatively pure strain? even to the point that the spore print drops heavily in the favor of the isolated genetics? I know this wouldnt be the case within the first few generations, I'm talking like 20-30 generations.
I have read that a strain will begin to lose its mojo after 3 generations (RR), but what if on the 3rd gen i do spore prints, then restart the isolation process, repeating the spore start every time the 3rd generation comes along? Will I eventually come down to a pure vialble strain, that sporulates the genetics the isolation was attempting? or will the genetics fail no matter what?
I have been searching for this, but haven't located the information that answers the specifics of my question.
