Time Constants On Metabolics

Hi everyone, I was interested in finding information about the exact biochemical reactions involved in psilocybe cubensis, in the process of tryptophan --> psilocybin conversion, specifically.

The pieces of information I am most interested in are:
-the time constants of the reactions
-the availabilities of each of the reactants

But any similar information links or pointers would be greatly appreciated.

Also, another question, are there other routes of reaction for the DMT in the mushroom? If it did not get utilized immediately is it possible it would break down?

I've posted on the topic a few times. You might want to search my username in advanced. You can also try adding "tryptophan decarboxylase" as a keyword to narrow things down.

It's almost certainly...
Tryptophan -> Tryptamine -> N-methyltryptamine -> N,N-dimethyltryptamine -> Psilocin -> Psilocybin

The reactions are decarboxylation, N-methylation, N-methylation, 4-hydroxylation, and O-phosphorylation.

Proper enzyme names, based on current understanding, would be...
tryptophan decarboxylase, N-methylase, 4-hydroxylase, and phosphorylase.

Kinetics data is usually not known for most biochemical reactions unless they are key parts of important pathways. It's not like they're happening in a flask that you can do one of those pain-in-the-ass kinetics experiments on.

> availabilities of each of the reactants

Availability of tryptamine seems to be the limiting part of the pathway. So production of tryptophan decarboxylase seems to be the limiting factor.

Not that I know of or can think of.

Probably not. I wouldn't expect it to be very reactive. It's usually broken down by monoamineoxidase (MAO), but I'm pretty sure there's none of that to worry about in mushrooms.

Thanks for the information fast, I'm happy to be able to learn so much about mushrooms here.

Do you happen to know if there has been any research put into triggers of enzyme production? As in, are there any established ways to stimulate or impede production of any of the enzymes, particularly 4-hydroxylase, and phosphorylase?

I don't know of any ways to stimulate production of any of the enzymes, but as with all biological systems nutrition plays a role.

Adding tryptamine has been shown to increase the actives. Otherwise there's no good ways to tinker with the system. At least not any easy ways.

Tryptamine or higher though, correct? (MT or DMT)
