Cloning mushroom question

Does cloning restart the number of transfers or does it continue on with the progression number? I have read 10 transfers for expantion on agar is the max number due to mycelium decline of vigor and vitality. This makes me wonder if the clone process resets the vigor trigger in the myc or is it just a continuation away from the mother culture? myc

A clone is just an aging of that line of cells, so it is a continuation away from the mother culture. You need to start with spores to "reset" the vigor I think.
I think the best option, once you find an isolate you like, is to create a master slant(s), and just start an working LC from it each time you want to work with the strain.

If you've started a culture from multispore inoculation once the mycelium has completed it's run and fruiting condition's have been established you will be able to determine which cake/casing produces the most mature mushrooms and then cutt tissue from the largest clusters that form on those cakes or casings.
After the tissue has run the agar, induce fruiting condition's and fruit straight from the jar to see which one is the most active/prolific/sizable and do as Psilocyentist explained and start a master culture slant or continue on with further isolation work.

I thought about asking this in advanced but lately people have been blasted by others for asking mundane questions.I am new to this hobby (9 months now). When I first started I didn't plan ahead or even think I would be getting into agar work. In that time I have used my original ms culture and now am left with one substrain which has been cloned several times just to see what would happen and it is underperforming. I wondeder if it had run its course? myc
