Long time for growth on Grain LC
    #13117842 - 08/29/10 09:34 AM (2 years, 8 months ago)

What would cause healthy grain LC to take longer to show signs of growth than MS?  Have 1/2 pint jars injected with grain LC from texas, guad, and GT.  Tex took off great, guad one little spot, GT nothing its been five days.  Innoced some birdseed, soaked only, 3 days ago with all three no growth on anything.  Have seen roger rabbit say 24hrs, have never seen this even with my fastest growing LCs.  I have seen some LCs take almost 2 weeks to show growth and be perfectly fine.  What causes this?  BTW 60 ml syringes with 15g needles were used with a shitload of myc in clear water.

Takes a few days for LC to start, usually. But not 5?

Thanks for responding Doc.  narrowed it down to a bacterial problem.  Would this cause the growth to stall but still eventually start?  This happened from not soaking the wbs in the master jars and the problem got into the LC i think.  The seed was soaked on the texas master jar and that one is working fine.  This failure really showed me how important it is to soak and rinse your seed first.  just always had problems with pooling even when drying on a towel for a couple hours so throw a lil verm in the jars now.  luckily theres a bunch of that texas and its the fastest ive seen besides brazilian.

How did you determine you have a bacterial problem?
You said you soaked the seed, but then you say you didn't?

Soak, simmer, strain to get best results.

Tell us how you made Grain LC, step by step.

made three grain LCs from 3 different fully colonized quart jars(1 tex, 1 GT, 1 GD) the tex wbs had been soaked no simmer and colonized fast, the GD and GT were just loaded and cooked or incubated at too high a temp cant remember exactly.  Then PCed a sealed quart jar filled with water with a silicone injection port.  wore gloves, mask, alcohol unwrapped 60ml syringes connected 15g needles rubbed alcohol on port sucked up water shot one per jar except two for the texas (luckily).  Then shook up pretty good and sucked everything back.  Flame sterilized between each jar.  the bacteria came from the high temps and long colonization times MS in quart can take awhile (terrible idea at this point in experience actually) or the nonsoaked wbs or a combo.  and its wet spot.

Things for next time
1.) use a smaller jar and much less grain for the master
any other suggestions?
