im getting ready to dunk some pf cakes. i noticed i left 2 too long and they have a few small mushrooms growing in them. can i remove those mushrooms prior to dunking and possibly use them to create a liquid culture. i realize i need to be cautious handling the cakes but im not new to handling cakes so im confident i can remove it without leavin any exposed substrate. can i isolate it and store it or whatever im at a loss from here>>

thanks for the input guys i appreciate it. i saw a tek where you could clone em to agar or pierce em with a sterile syringe then take that section and put it back in solution. would it be better to take em off after i dunk em since they're hydrated more or does that even make a difference?

take off before. your just going to dry them anyways and yes you could clone if you wanted to.

This is an awesome resource. Thanks again for the help. I'm new to the shroomery but was introduced to the PF tek back in '96, when all you had was an instuction booklet. It was sink or swim!! So i've been out of the loop for over a decade, but this is my first attemt at spawning to bulk (coco/verm/gyp). I chose brown rice as spawn since it was familiar. Next time i'll knock up rye with a liquid culture!

Yes you can remove these mushrooms prior to pinning.
Do not use your small pinhead mushrooms to clone to a liquid media.
Your small pinheads contain contaminates. Those contaminates will destroy your liquid media with contaminates.
Clone small pinhead mushrooms to agar. Any contaminates can be cleaned away when you place pinheads to agar. Some times even out running any contaminate mold spores.
Do not clean healthy mycelium from contaminated agar culture in front of a flow hood. Cleaning your agar culture in front of a flow hood will blow the contaminate spores all over your lab.
Clean contaminated mycelium form contaminates in a clear plastic trash bag. Preventing the spread of contaminate mold spores through out your house and grow shows.
Isolation of strains. Obtainable on agar.
Use live agar culture wedges to inoculate liquid media.

wow that was informative and you answered my agar question. In regards to isolating contams I saw that on the "Lets grow mushrooms" dvd to just remove a grain size section of the viable myc from the agar dish if it showed contams and place that in a new agar culture dish and go from there. So since I dont have petri dishes I'll improvise. I saw a couple teks like the grocery store agar teks or something like that. appreciate it!

Readily available. Pint size wide mouthed jars work well for agar.
Only pour a small amount of agar.
Fill your pint sized wide-mouthed jars with 1/8th to 1/4 an inch of agar media, per jar. Mycelium only colonizes on the surface of agar. Not through the center.

hamloaf i saw your link for grain prep, i used that same tek to the letter! i knocked up rye berries with pf classic along with some pf style cakes, the rye berries are moving slow, i think one cc of multispore per quart was too week....any suggestions? Im gonna clone a mushroom from my first flush. i plan on transferring some to agar, as well as a liquid culture via spore print. im working with pf classic, b-plus, and keepers creeper. can you give me any input.....

is there a difference in the gypsum i bought two different kinds one i bought at a beer brewers and its fine white powder the other i bought at farmers co op and its grey with black flecks and is used for lawns. i was just wondering if the grey is okay before i throw it in there

These cakes were cased with 50/50 peat moss/verm, 10% calcium carbonate, and 10% gypsum on the 29th then incubated until the 5th. One half-pint pinned 2 days after going into the fruiting chamber. After five days I picked 4 shrooms (over 60 grams wet), and still has 2 more on the side and a number of small pins. I did notice that the casing itself has shrunken. The second half-pint tray had a 1" casing layer and is now just developing small pins. One mushroom was as round as a quarter in diameter in terms of the stem and the cap was about 3 1/2 by 3 1/2 and weighed 35 grams wet. I am currently printing the cap and cloning the stem via agar. This strain is Keepers Creeper!

i use half pint jars.the lids have RTV silicone at the injection site for self healing and a second hole for gas exchange covered with a tyvek filter and sealed with RTV. I basically use the grocery store agar tek with a few modifications.look it up and private message me with qestions if you want.they also have teks for lid prep with RTV look them up cause its cheap yet effective. as far as the agar glass petri dishes are the best im going to order some soon.those RTV lids are also great for rye berries or WBS shit like that.

Heres some pics of keeper's creeper on BRF crumbled and cased with peat/verm/cal carb. I got a lot of pins and about 7 grams cracker dry. I have dunked them 3 times for just a few hours in water/peroxide then re-cased with very thin casing layer. The next few flushes ranged between 2-4 grams. Overall, I easily netted over a half-zip bone dry. I just dunked it again for kicks.
