When it comes to making LC, less is better.  The fewer spores you have the fewer competing strains you'll end up with.    Technically you only need 2 active, compatible spores to create an LC.  In any given spore syringes you'll probably have tens of thousands.

I don't have the necessary workspace or tools to do this, but if I did here's how I would do it.

1.  Start with spores on agar.  Isolate out 10+ strains.

2.  Create an LC with each isolate and do 1 cake of each.

3.  Keep the one(s) with the best fruiting strength/potency (start over if they all suck).

4.  Store the original LC's as a master culture.  Using literally DROPS of it to start new LC's when needed and using those to fruit.

Doing it this way you'd have access to an infinite amount of inoculant that fruits well and is potent.

However since I don't have access to the necessary tools atm, I'll settle for making LC's from spores and doing it that way :-D

Using injection ports  tyveks I've had a very high success rate with LC's.  Always do a test inoculation before going wild.

I've used both Lme/Dex and just LME.  They both work about the same so I don't see the point in adding the dex anymore.

I haven't had any problems with the LC's not taking to grains or anything though.

Thanks so much spongiform! I Live a couple of miles from a home brew supply store and I will look for the exact brand that you linked to amazon. I am confident that I will find some LME there and I won't have to wait for shipping!

Please correct me if I am wrong I need 1 quart boiled distilled water.
Add 1 Tablespoon LME and let dissolve. Pour 4oz into each of eight clean 1/2 pt low profile canning jars with 3 ply tyvek lids and a 1/4 inch blob of silicone on  both sides of tyvek to make a self healing injection port. cover with Al Foil and PC at 15 psi for 20- 30 min. Allow to cool in the unopened PC till they are room temp. Inoculate them with 1/8-1/4 cc of spores in my glove box. give each a swirl and store in a warm cupboard for a few days gently swirling several times a day till they thicken?  Sorry if I sound so ignorant...but I am..and you Sir are a wealth of experience and knowledge:mushroom2:

Pretty close.

Here's how I do my lids:



2, 1/2" holes one with injection port and one with tyvek glued over it.  I use a little bit of silicone to make sure the port is air tight around the edges.

You can do the same but make your own injection port.  Put a good quarter sized blob over the hole. 

After dissolving the LME via boiling you need to filter it to remove sediments.  I accomplish this by just running the solution through a coffee pot with an empty filter in place to catch the crud.

You could probably rig up something similar with a kitchen washcloth or papertowel.

After that load into jars and PC.  Foil is optional.  About the only thing is accomplishes is keeping dust/airborne crud from landing on your lid before inoculating.

When inoculating I use an alcohol lamp + alcohol wipes (papertowel + alcohol works).  First wipe down the port with alcohol and while that sits flame heat the syringe red hot then cool down with alcohol wipe and keep it covered until you're ready to inject.  Make sure the syringe is shaken up a bit so the spores are spread out.

After inoculating do not shake.  Let it sit about 3 days then start to take/swirl.

This gives the spores time to germinate and make contact with other spores and begin their process.

After take give it about a week at room temp, swirling occasionally or set up a stir bar system. 

I recommend 20-60cc syringes with 16 gauge needles for drawing up the solution to inoculate.  Follow the same sterile procedures for this.

Just curiosity Spongi, but you say that 1.5g's hits you and your buddies pretty hard with the APE's.  I am wondering how much it generally takes you to reach the same level with regular cubes?

1.3g was roughly equal to what I'm used to feeling with an 8th.

Seems with pe you have less confusion and more visuals.

I'm more aware of my surroundings and what's going on but everything looks crazy.
