so, did you autoclave thiamin separately at pH below 5.0 as you point out here?

Hi Speeker,
No, I only add about 50 mg of thiamine hydrochloride solution in every flask and autoclave everything together. I did not measure the final pH. The thiamin hydrochloride was obtained from a medicine that contains 300 mg of thiamine hydrochloride per tablet. I prepared a solution dissolving one tablet in water and used a enough quantity in order to have about 50 mg in the potato broth (35 ml of potato broth/flask).I also replaced the white rice for brown rice (more vitamins).
Cheers,
Wilson

Nice job! I've been gone for a while, and I'm staying away from this site for a little. Glad to see that you put that strain to use

A few more pictures using the Roger Fudd´s Cordyceps militaris strain . The first picture is from the first test and it is showing all the mushrooms covered by mycelium. I think this happened because the mushrooms were harvested too late and it could also be a sign of lack of aeration and/or excess of moisture. The dates that appear in the pictures show when they were taken

I was expecting a formation of spores, as shown in this wild specimen

But some different structures like cracks appeared.

The two pictures below show the formation of primordia on the surface of the mycelium grown in liquid culture:

The three photos below show some cultivated mushrooms in a medium different from that used in the first test. I used a different formula with the addition of silkworm pupa powder. The mushrooms appeared in smaller amounts, but they were more robust. The environmental temperatures varied widely from 13 to 28 º C within the same day sometimes. I think this was also a big problem.

Apparently the first test worked better than the second. More tests will be needed and a better environmental control too.

Wilson

This is a summary of the recipe and the methodology used on my first experience with Cordyceps:
Cook peeled and sliced potatoes in mineral water or distilled water on low heat until tender. Filter the broth through a sieve and discard the potatoes.
Potato Broth
500 g potatoes (peeled and sliced thin) 5000 ml water (distilled or mineral water)
Prepare the following nutrient solution using the potato broth prepared above:
Nutrient Solution
200 ml potato broth 2 g KH2PO4 1 g MgSO4 1 g C6H14N2O7 (Ammonium citrate) 30 g glucose (dextrose) 3 g Soybean peptone 50 mg vitamin B1 (thiamine)
Dilute all the above components with water by adjusting the volume to 1000 ml
Measure 35 ml of nutrient solution and pour into wide mouth glass jars. Weigh the rice grains (30g) and put them in the jars along with the nutrient solution. Close the glass flask with a metal lid with a felted disc under the lid. You do not need a hole in the metal lid, only the filter under the lid is enough for the exchange of air during the growth of the mycelium.
Per jar :
35 ml nutrient solution 30 g rice (polished or brown rice)
After sterilization, inject approximately 5 ml of the Liquid culture of Cordyceps militaris into every glass jar (use a micropipette with sterilized tips or even a stainless spoon sterilized on the flame of a Bunsen burner). Incubate in the dark (25ºC) until the mycelium cover all the grains.
Primordia initiation
Start a one week treatment for the initiation of primordia by putting the flasks on the refrigerator at night and leaving them at an illuminated area during the day (18-25 ºC), without direct sunlight. After the appearance of primordia , keep the flasks in a 18-23ºC environment with indirect light until the mushrooms develop to full size.
You can find more details about the environmental conditions on the following translated link (indicated by Speeker) :

I emailed them and they said possibly but they were afraid I might compete, and I am an honest man and I never know the future, but of course I would never try to sell their patented strain, but I am just wanting a basic cordyceps sinensis strain, who knows maybe I can get me some rattlesnake venom and play around myself Being in college may be I can play with a hplc. Anyone with a cordyceps sinensis strain that is willing to share, please?
