Tissue to plate, then best sector transfered to these new plates. You guys see any more sectors in there, or do they look isolated?

Sample was taken from stem and put to agar. Grew that out, took best sectors from those plates, and put those to fresh agar. Looks good to go to grain, then?

Yes. You'll rarely see sectoring with clones. When spores are swiped on agar, you'll often have hundreds of sectors develop.

differences between strain isolation by transfers and by cloning?

Okay.. I was just making sure. If you have more than one strain per fruit will you not necessarily see the sectoring on the plates? Two plate generation with tissue clone is pretty usual over the multi-generational spore to agar? I just don't have the time to do all the sectoring involved with the spore->agar route, and if I can save the time, plus have a heads-up on what I may end up with, that's all good for me...

My clones will start pinning on the agar without transfering.Ill take a peice of that and throw it into a malt/dex lc.

I could be wrong but it looks like bacteria all around the perimeter of those plates.

Where do you see this? I see the reflection from the flash on the wood grain. I see myc. I see condensation from the lid of the agar. I don't see wet rot. Enlighten me.

well to me I see the flash as a white gloss, along the left side and bottom of the lower pic there is quite a bit of what looks to me wet spot contamination, maybe the defone would be able to clear it up? Asshole-

To me, it seems more like water between the dish and the surface. But I could be wrong, too.

I see bad advice here all the time. People take that stuff to heart. I wouldn't want someone tossing out a perfectly good agar plate on someone's random diagnosis. You can call me an asshole, but he didn't ASK if it was contaminated. Just if it looked like an isolated strain (which it does).

And, yeah, you put "I could be wrong." But this is in the ADVANCED MYCOLOGY forum. I think most know what contaminations look like. Especially someone who has the agar plate in front of them and is NOT asking if the dish is contaminated.

Just watch the advice you give people. There are a lot of noobs on here, people doing their first grows, or just people looking for GOOD and VALID information.

Just my 2 cents

That was a light hearted "asshole", not specific to you who I have no grounds to form an opinion on, but a universal "asshole" sent out to an attitude because I see bad protocol on these boards all the time, the Shroomery has a particularly bad reputation these days and is known throughout the OMC as being antagonistic and as a result, "not what it used to be" Kind of like when a great musician becomes arrogant and egotistical because of there sucsess than leaves their band only to suck. I didn't advise him to do anything. And yes I considered that he was sectoring which would lead one to believe he/she is experienced to some degree. But for all we know, he/she busted a PF cake once and is isolating a phen from that and has never seen bacteria on agar, which IME can actually just look nutrients that have surfaced if its a homemade agar plate.

If I add a suggestion, that someone might want consider taking a look at some questionable growth before contaminating 38 qt jars and waisting the time/money it takes to prepare them, I think as far as advice goes, thats pretty fuckin' sound.
