wtf is this sh*t in my agar

heres my pan cyan agar plate...
as you can see theres a load of nasty crap in there...
and a few white spots of pan cyan mycelium..
what is that

this is common for me with agar...but for some reason i still stick to it
rather than lc's :crazy:

Anytime you have agar open to the air like you do above, it will fill up with bacteria and whatever mold spores are floating around at the time. You'll need to work in a glovebox or in front of a flowhood, and never have the lid off more than a few seconds while making the actual transfer. You need filtered gas exchange too. The depth of jars encourages stale air in the bottom, favoring contaminants. Petri dishes do not have that problem.

Alder is not the BEST type of to use
Heat drying cubes is okay and recommended
Sunflower seeds do not cause contams
You should directly mist your cakes

The subjects that I cover are not species specific and can be used in All Aspects of Mycology

That's a lot of Bacteria growing.

You should not move on to work with LCs until you've mastered agar, since you won't be able to see the contaminants nearly as well with LC.

i saw that you can extract this mycellium from a petri dish to another so there are less and less contaminant each time ...

Yes, that is called transfer. However, I think wutang needs to work on his technique before he will be successful with clean transfers. And get some petri dishes!!

no thing is i've had some live cultures before then i moved to agar ,
agar is easier for me
i dont think theres a certain order that needs to be followed.
but yeah i do need some dishes

cut out the good stuff and keep jumping till you got what you want
then jump for rihizos
at least thats how i do it, and its fun

yes being sarcastic.
1/2 pints can work well but you need to let them breath.
put a polyfill filter in the lid or use them upside down so that they do not seal

Get your jars and fill with agar and then p cook them, pouring agar into after its cooked will only increase your contam possibilities.
If you choose to use them with upside down lids then give them a day or two to cool off so that excess moisture that forms on the lid will dry. This moistur will cause a slight seal and removing the libs will suck outside air in.
Never open those things in the open air.
Make a ghetto glove box. Get a clear rubbermaid container-fc and flip it over and cut two holes in it for your hands.
Put everything in there and spray the hell out of it with alcohol. Work inside it and limit the time you leave the lids off the agar. Dont use a flame inside it unless the alcohol has disopated.
get your first flush of mushrooms and use the money to buy some glass petri off of ebay

the success rate is significantly low without the gb when cloning.
it happens but most of the time it gets bacteria and then finaly theres a dish
thats fine..but its cloudy mycelium rhizo

cultivation is easy without a glove box or hood.
The pf tech only takes a house with somewhat livable cleanliness.
Working with agar only takes a bit more cleanliness, cloning gets tougher.
Working with a good glove box or hood is awesome once you have done so without
