I was hoping someone could describe or post a picture of what a mycellia inoccula should look like before use. Recently inocculated Sabouraud's media w/Eq spores: after 7 days at 30 C there is a nice cloud in the media. I have taken 50 uL samples at 2 day intervals, heat fixed, gram stained and looked at under 100x and see NO SIGNS of contamination; therefore I believe that the little cloud is 100% mycellia. How thick will it get? should I just shake the bottle hard (100 mL) to break up the cloud and use 1 mL per injection point? Please provide some advice. Thanks in advance
yes, you are in good shape. you do not need a huge cloud just a nice solid one. when you shake that fellow it will cloud up the whole jar.
and shake it really well to break up the myc and then pull out however many syringes you need.
as how much to use per jar, i would say it depends on the type of work you are doing. if pf jars then 1ml will be plenty per innoc point. on the other hand if you are working with quarts of grain then it seems 3 to 5ml works best per jar.
